Oat belongs to genus Avena L. in Gramieae. It is an annual crop with high feed andfood value. Different explants, culture medium, genotype, hormone type and concentrationratio from Bai Yan No.2, Gansu No.3and Denmark444mature seeds were studied,influence factors of callus induction, following generation and differentiation were explored,the high-frequency of regeneration system from oats has been established. The resultsshowed as follows:1,Effects of the callus rate on the mature embryos, hypocotyls, young leaves werestudied, the results showed that: the callus induction rate was the highest,76.11%,and thecallus state was optimal on the mature embryos as explants.2,Comparative tests on the MS, MB, N6basic medium found that the oat callusinduced effects best on the MS basic culture medium, the induction rate was the highest,40.74%,compact callus and fast growth.3,With sterile water, CaCl2,4℃low temperature and PEG on the oat seed pre-treatment,results showed that seed pretreatment soaked in sterile water was best, the method is simpleand the effect is optimal.4,There is a very significant effect on the scutellum placed way for the callus inductionrate, it s best to the callus induction on the way that the oats mature embryo scutellumdown direct contact with the medium, the callus induction rate increased by21%.5,Interactions existed between genotype and culture medium, callus induction rate wasdifferent with the different genotypes in the same medium, there were also significantdifferences with a genotype in different media, callus induction rate was the highest on the8thtreatment of Bai Yan No.2and Gansu No.3,63.33%and98.33%respectively,forDenmark444, the highest callus induction rate was on the1stand6thtreatment,100%,thecallus from the6thtreatment was non-embryonic, can't differentiate into embryonic seedling.6,Cytokinin and auxin ratio of3:1is appropriate for the callus of BaiYan2,differentiation rate of Gansu No.3and Denmark444was the highest with the Cytokinin andauxin ratio of2:1,52.38%and61.54%, respectively. 7,The hormone type and concentration proportion were researched to screen the bestmedium for the callus induction and subculture, suitable medium for callus induction frommature embryo in Bai Yan No.2and Gansu No.3was MS+4mg/L2,4-D+1.0mg/L NAA, forDenmark444,the suitable callus induction medium was MS+2mg/L2,4-D, subculturemedium was MS+1.5mg/L2,4-D+1.0mg/L NAA+1.0mg/L6-BA.8,The best differentiation medium were screened for the different genotypes of oatsthrough the system research, suitable medium for callus differentiation of Bai Yan No.2wasMS+0.4mg/L NAA+0.2mg/L KT+1.0mg/L6-BA; for Gansu No.3the suitable callusdifferentiation medium was MS+0.8mg/L NAA+0.8mg/LKT+1.0mg/L6-BA,andMS+0.2mg/LNAA+0.4mg/LKT+1.0mg/L6-BA was the suitablecallus differentiation medium for Denmark444.9,Study on the regeneration frequency showed that, regeneration frequency of GansuNo.3was the highest in three genotypes of oats, about61.56%, can be used to establish theregeneration system of oats and further genetic transformation.
