Deoxynivalenol (DON), a.k.a. vomitoxin, is a mycotoxin mainly produced by the fungi Fusariu culmorum and F. graminearum while growinig on crops. It is a trichothecene with relatively low toxicity. However, DON is of interest because it is one of the most common contaminants of wheat, corn and barley and only partly reduced during processing of grain. In the present experiment, five different concentration of DON were adopted to induce PIEC, HUVEC and Lovo cell lines for 24h. Then the morphological changes of the cell lines were observed under microscope. The MTT method was performed to determine the inhibition ratio on proliferation of PIEC, HUVEC and Lovo cell lines. And TUNEL method was used to assay the apoptotic rate of these cell lines. Immunohitochemical method was employed to measure the expression of Caspase-9 and Bax in apoptototic PIEC, HUVEC and Lovo cell lines. The results showed that different concentration of DON can inhibit the proliferation of these cell lines and higher concentration can change the morphology of PIEC, HUVEC and Lovo cell lines. The inhibition on proliferation of PIEC cell line is the most significant out of these three kinds of cells compared to the controls (P<0.05). Different concentration of DON induced apoptosis of all the cell lines, for PIEC and Lovo cell lines, the apoptotic ratio reached the highest level at 500ng/ml (71.74% and 60.32% separately ), and HUVEC at 5μg/ml (26.61%). Then the apoptotic ratio decreased meanwhile the concentration of DON increased, which may result from the collapse of apoptotic cells and their falling from the cell slice. Besides the result implied that the Caspases pathway may contribute to the apoptosis of PIEC induced by DON, and the inhibit gene Bax may promote the process of the apoptosis. The changes of Caspase-9 and Bax were undetectable in the process of apoptosis of HUVEC and Lovo cell lines, so we made a conclusion that the process did not relate to the Caspases pathway and gene Bax.In the present experiment, baicalin, dehydroandrographilide and aspirin were adopted to induce BGC-823, HCT-8 and Lovo cell lines for 48h, 96h and 1w.
