Studies On Shoot Tip Tissue Virus-free Culture And Rapid Propagation Technology Of Grapevine

This paper built an easy and efficient grapevine stem cultivating and detoxification technology system by discussing the detoxification technology of grape virus diseases and studying the rapid propagation technology after detoxification, and screened the matrix suitable for the grape to grow by screening the cutting medium of the summer black seedless grape, so as to provide the most suitable matrix for the acclimatization and transplantation of the virus-free plant. The technology of rapidly propagating the virus-free plant after the detoxification of the grape was discussed. The following research results were mainly gotten:(1) Four detoxification methods by combining the stem tip culture, heat treatment, chemical treatment and heat treatment, and chemical treatment; the detoxification effect of the combination of the heat treatment and stem tip culture is better than that of separate stem tip culture, wherein the detoxification survival rate and detoxification rate of the variable temperature heat treatment was higher than that of the constant temperature heat treatment, the comprehensive detoxification efficiency of the combination of the heat treatment and chemical treatment was the highest, but the survival rate was low, the operation time was long, and the growth and development of the stem tip was slower than that of other detoxification methods. The most suitable detoxification method as to detoxify by combining the variable temperature heat treatment with the stem tip culture.(2) It determined that the best sterilization treatment scheme was that lmol/L 30s hydrochloric acid+1% sodium hypochlorite treatment for 20min has the best sterilization effect of the single-bud stem segment of the grape; the medium of the best primary culture, subculture, rooting culture and stem tip culture of six kinds of grapes, i.e. summer black seedless, golden finger, Hutai No.78, Venus seedless, Yongyou No.l and Ruby seedless, was screened; and the tissue culture and rapid propagation system of six kinds of grapes was built.(3) The comprehensive effect of the quality of the summer black seedless grape cutting seedlings was the cutting seedling by mixing river sand:edible fungi residue:grass carbon (3:2:1), the quality of nursery stock cut in the signal matrix river sand was the poorest, and when shoot cutting with the virus disease was stored and the virus-free seedling was transplanted, the formula of river sand:edible fungi residue:grass carbon (3:2:1) shall be selected.
