Garlic (Allium sativum L.) is a perennial herb of the Liliaceae Allium L. Garlic bulbs, stems andseedlings can be consumed and it is an indispensable spices and vegetables in our daily life.Garlic in the production of asexual reproduction, vulnerable to virus infection led to declining the yieldand quality, making Xinjiang garlic acreage decreased year by year and has seriously hampered thedevelopment of the garlic industry. The application of heat treatment and garlic shoot tips tissue culture isan effect method for virus-free. Garlic shoot tip culture is difficult and low propagation coefficient, thehigher cost of production of virus-free species becomes the bottleneck problem of the garlic virus-free seedcultivation and production applications.In this study, used heat treatment combined with bulbils and shoots tip tissue culture removal of garlicvirus and used induced adventitious shoots garlic bulb disk as fast reproduction. Study the effect ofdifferent hormone for bulbils budding bulb shoot tip callus and adventitious shoots, bulbs disk adventitiousshoots induction and rooting of garlic. Study the different shoot tip size and detoxification effectrelationships and virus-infected garlic and healthy garlic physiological structure. Following showthe result:1In Xinjiang white garlic and Yining red garlic bulbils germination, the highest germination rateof300mg L-1 GA3concentration, performance was significantly2used heat treatment and shoot tip tissue culture establish garlic shoot tips virus-free culturesystem. The results show that, MS+0.3mg/L NAA+1.7mg/L6-BA conducive to Xinjiang whitegarlic and Yining red garlic shoot tips adventitious shoots. Stripping shoot tip size of0.5mm,Xinjiang white garlic and the Yining red garlic callus rate were70%and80%, the adventitiousshoots incidence were60%and70%.3The experiment adopted quadratic regression orthogonal rotation combination design and DPSprocessing the number. Xinjiang white garlic and the Yining red garlic bulb disc adventitious shootsgrowth hormone best ratio were: MS+2mg/L NAA+7.5mg/L6-BA and MS+1.799mg/LNAA+7.5mg/L6-BA. Under these conditions, the Xinjiang white garlic and Yining red garlic bulbs diskShoot Growth maximum value of39.22and22.92. Xinjiang White garlic and Yining red garlic rootinduction best formula is: MS+0.5mg L-1IBA+1.0mg L-1NAA.4Infected garlic leaves from non-infected garlic leaves compared to leaves rolled inwardsdistorted, and can not stretch properly, showing deformity. The mesophyll palisade tissue thannormal cells arranged in loose leaves and most irregular shape, spongy tissue cells infected heavierleaves becomes loose expansion disordered.5Better to0.5mm size of shoot tips detoxification effect of the garlic bulb stripping shoot tipvirus-free, virus-free rate of up to100%. Bulbils bulbs disk tissue culture also has a detoxificationeffect, Xinjiang white garlic and Yining red garlic detoxification rates were58-86%and54-85%.
