In order to explain the relations of Rosa cultivars in Qingdao district, palynology and RAPD analyses were made. The results are as followings:1 In this paper the pollen morphology had been observed with electronic microscope. The conclusion displayed that the pollens of Rosa were very similar in morphology. Most of them were super long or long sphericity . They were oval or long oval in equator view, 3-labled-rounded or subrotundity in polar view. And 3-colposate with pore film, the style of pollen is N3P4C3.The polar axis of the pollen grains was 32.26μm—49.μm,and the equatorial axis 16.20-23.15μm,which belonged to middling.2 The rosebushes for experiment were classified into 4 categories based on cluster of palynology.I R.multiflora 'yinhaifanxing' , R.multiflora 'hongguoyudie' , R.multiflora ' yinpanjinxin' , R.multiflora 'bingdie', R.multiflora 'jiaorongsanbian', R..multiflora'duiyun'; II R.multiflora'rushuang', R.multiflora var.cathayensis, Rmultiflora 'bingqing' ; R.multiflora 'danfen' , R.multiflora 'fenxia' , R.multiflora 'yanzhi' , R.multiflora 'qiaofen' , R.multiflora 'hongtan' ,III R.multiflora 'fenmiantaohua' , R.multiflora 'huanyan' ,Cl.President L.senghor, Cl.White Masterpiece, Cl.New Dawn;IV R.multiflora 'yudie' , R.multiflora 'diewu' .3 DNA of Rosa multiflora was extracted from the late-winter buds and the early-summer leaves. The optimized reaction system and program of RAPD for Rosa multiflora were developed.Random amplified polymorphic DNA(RAPD) was used to analyze the genetic polymorphism of Rosa multiflora cultivars. Sixteen primers amplified 175 DNA fragments, 151 (86.3%) of which were polymorphic.4Cultivars were taken for cluster analysis by the way of UPGMA. When LD was 0.667 the material could be classified into 2 categories. I R..multiflora 'yinhaifanxing' , R..multiflora 'rushuang' , R..multiflora 'yinpanjinxin' ,R..multiflora 'bingdie' , R.multiflora 'yudie' ,R..multiflora 'diewu', R. R.. multiflora' jiaorongsanbian', R.. multiflora' duiyun', multiflora' hongguoyudie', R. .multiflora' qiaofen', R..multiflora var.cathayensis, R..multiflora 'bingqing' ;R..multiflora 'danfen' ,R..multiflora 'fenxia' , R..multiflora 'yanzhi', R.multiflora 'fenmiantaohua'; II R..multiflora 'huanyan', Rmultiflora 'hongtan', Cl.President L.senghor, Cl.White Masterpiece, Cl.New Dawn. 5 The cluster of RAPD were almost unanimous with palynology on the whole, but there were also some difference . The I II(R..multilora'hongtan' excepted)of palynology cluster accorded with the I of RAPD cluster. The III (R..multiflora 'fenmiantaohua' expected) of palynology cluster accorded with the II of RAPD cluster. Combined the cluster of RAPD with palynology and morphology, the results will be more credible. It can offer some instructive help to Rosa cultivars breeding and conservation of germ plasm resource. And how to applying three of them into cultivar classification of flower still need to be researched.
