The quality improvement is one of the important object for wheat breeding. Wheat include protein,starch and fat.Theeffects of the protein quantity and quality is the most important with respect to wheat quality properties.The component of wheat protein that include glutenin and gliadin is responsible for wheat dough quality. Glutenin subunit consists of high molecular weight glutenin subunits(HMW-GS)and low molecular weight glutenin subunits(LMW-GS).Its allele subunits have abundant polymorphism and their alleles compositions are important factor on dough quality. Recenly,the few report of glutenin quantity and bands are both in abroad and China, there are more LMW-GS which are still disputed on its nomenclature and function on quality because of test material and mothod.The 188 Chinese wheat mini-core collections were analyzed using SDS-PAGE for gaining its composition of HMW-GS, over expression of Bx7,its subunit of Glu-A3 locus and bands.Quantitying of glutenin was analyzed by gel imaging system and Quantity One Software,at the same time, SDS Sedimentation Value was measured and the correlation was analyzed.The three special wheat varieties were analyzed using SDS-PAGE for gaining the protein output in Glu-A3 locus.The result showed as follows:1.The 188 Chinese wheat mini-core collections were screened by SDS-PAGE. The results showed there were 16 alleles subunits and 15 unknown subunits at Glu-1 locus.The 36 subunit compositions were identified at Glu-1 locus included N,7+8,2+12 were the highest.The average quality score of the varieties were 6.4,23 cultivars over expression of Bx7 were detected,the quality of over expression of Bx7 was more than normal expressions,the patterns of 2~*,7+8,5+10 were the best,the worst of N,7+9.2.The number of c and a subunits were 39.98%and 25.00%,e subunits were 4.79%,the quality of d was the best,the worst of e subunits.The bands analysis of LMW-GS showed,for the B zone,24 bands were detected,which formed up to 28 different patterns,for the C zone,22 bands were detected,which formed up to 20 different patterns.3.The three special varieties were analyzed by SDS-PAGE,the result showed the molecular weight of their variaties were different from the seven allele subunits of the seven standard varieties in Glu-A3 locus,so we think there were new proteins in Glu-A3 locus in these three varieties.There were 3 subunits types which were 1,2~* and N at Glu-Al locus of their three varieties,the quality score were all 6.4.The quality analysis showed the coefficient of variation in C area glutenin was the largest,glutenin was the smallest,the correlation coefficients of SDS sedimentation value and glutenin was 0.51397.SDS sedimentation value and quality score in Glu-1 correlated to the quality of glutenin,A area,B area and C area.
