| Castnopsis sclerophylla Lindl.is the kind of arbor,which is abound in the southern mountain and Dabie Mountain area of Anhui province.Castnopsis act as one kind of defensive woods for its special structure of wood.Its fruit can diminish inflammation and has anti-bacterial effect,and is edible food,according to Chinese traditional medicine records.To promote the mountain agricultural structure adjustment and economic development,promoting income,it is necessary to research this mountain plant,which researched less in the past.In the paper,the paste properties of Castanopsis powder have been studied with modern instruments such as polarizing microscope,spectrophotometer,rotary viscometer etc.The results showed that Castanopsis powder has high gelatinization temperature (65.5℃~78℃),lower clarity,better retrogradation and bad freeze-thaw stability.In the pH value range 6.0-8.0,Castanopsis powder paste has higher viscosity,excess acid or alkali changing viscosity.Temperature and rotate speed have some effects on paste viscosity properties,has the "shear thinning" phenomenon.Castanopsis powder consistence has obvious effects on paste viscosity properties,when powder consistence over 5%paste viscosity hoiked.According to detect food ingredients ministerial standards,the protein content,fat content,mineral content,total sugar content of Castanopsis powder have been measured with Kjeldahl,Soxhlet extraction method,atomic absorption spectrometry,3'5' two nitrosalicylate method ete.The results showed that its protein content is 4%;Crude fat content is only 1.73%,and have high content of minerals,the human needs of conventional metal elements have higher content in Castanopsis powder,its heavy metal content analysis showed that the powder Castanopsis suffered less pollution.The total sugar of Castanopsis powder is 91.4%that showed it is polysaccharides food.Castanopsis polysaccharides,the efficacy-basis of Castanopsis powder,have been extraction and purification using following steps:degreasing,then wiping off soluble polysaccharide with distilled water repeated washing,and dispersion with lye solution, and further separation using butanol-isoamyl alcohol,then depositing with ethanol, centrifugation,and last wiping off protein with salting in for it is water-insoluble.By alcohol ractional precipitation showed the prepared Polysaccharide is a single component. Its' protein content is trace by mensurating.Using chemical analysis,show sulfur degree of substitution of Castanopsis polysaccharide is 0.28.The degree of substitution is lower than which the reported sulfated polysaccharide which chemical modification prepared.Using Fourier transform infrared spectrometer detection showed that sulfate absorption peaks are 1240cm-1. 850cm-1.It show sulfate group is conjunction with sugar ring,and it also demonstrates the sulfate group of Castanopsis polysaccharide is a combination of biological.NMR spectra show that the Central C chemical shifts of Castanopsis polysaccharide is similar to reported[q -Glc(1→3)-]n.And from the spectra can be seen in 68.469ppm have weak signal,further confirmed sulfate groups on the C-6 position.To summarize the above,by the powder Castanopsis nutrient content analyzed,and the main ingredients Castanopsis polysaccharides structure analyzed,sulfate groups in the degree of substitutionmensureted,sulfate group to replace the position analyzed showed Castanopsis powder is a new functional food resources with certain physiological effect.The structure of Castanopsis polysaccharide is[q -Glc(1→3) -]n,and about every four glucoses have one is modified by sulfate group on its C-6 position.The research about Castanopsis polysaccharide in this paper is the first of its kind in China.Research analysis of the methods used,such as alkali-soluble extracting, salt-soluble wiping off proteins and direct measurement of solid samples magnetic spectra etc,can provide reference for following research about Castanopsis polysaccharide. |
