Castanopcis Sclerophylla(Lind1) Schottk,also known as Castanopsis chestnut,Castanopsis tree and so on,is evergreen arbor belonging to the branch of cupule. Castanopcis Sclerophylla is one of the main populations of broad-leaved forest in subtropical zone.The medicinal values of the fruits of castanopsos sclerophylla are quite high.According to "Compendium of Materia Medica" introduction,the main treatment of the fruits of castanopsos sclerophylla is to cure Impotence,Edema,and it has functional effects of profiting breath,allaying our hunger, brighting the eyesight,strengthening the bones and muscles,assisting male genitals, repairing weak fatigue,strengthening waists and knees,covering colour,and so on. In the drainage area of FU HE River in Jang Xi Provience,it usually used to cure Diarrheal diseases.The paper made the drying fruits of castanopsos sclerophylla as the raw material,considered from the comprehensive utilization of resources,we adopted the High-tech of Natural Products Extraction and Separation,such as Smash technology,Ultrasonic extraction technology,Microwave extraction technology, Macroporous resin separation and purification technology to extracting,separating and purificating total flavones from Castanopsis Sclerophylla.The properties of anti-oxidation,anti-bacterial were also studied in this paper;preparing the Sclerophylla pectin by acid extraction,ethanol precipitation;isolating a high level of Sclerophylla starch from the fruits,and also carried out the basic research for its properties.The main contents of the paper and the results are as follows:1,The study of the optimal technology of Ultrasound-assisted extracting total flavone from Castanopcis Sclerophylla(Lind1) SchottkStudies have shown that there were good results with ultrasonic technology to extract Castanopcis Sclerophylla(Lind1) Schottk flavonoids.On the basis of the ultrasonic time,ethanol concentration,the ratio of solid to liquid,the numbers of extraction as single-factor,by orthogonal experimentation we got the optimal extraction condition of ultrasound-assisted extracting total flavone from Castanopcis Sclerophylla(Lind1) Schottk is the Ultrasonic time 65 minute,ethanol concentration 40%,solid/liquid ratio 1:30,and the number of extraction were 2 times.A colour reaction and UV scan were studied of the extracing solution,and then the flavone from Castanopcis Sclerophylla was elucidated as flavonol.2.The study of the optimal technology of purificating total flavonoids with macroporous resinThe Experiment used five kinds of macroporous resin named AB-8,D-101, DM-2,DM-130,NKA-9 as the filler,and discussed its adsorption performance, desorption performance,etc.By Studying,we got there was a good effect of purificating Castanopcis Sclerophylla(Lind1) Schottk flavonoids with the AB-8 macroporous resin.The optimal purification conditions was that:the suitable adsorption pH was 3; and the right conditions of eluting was as follows:ethanol concentration as 40%,the flow rate as 2BV/h.In these conditions,the macroporous resin had good absorption and desorption properties;the AB-8 resin's static adsorption reached more than 2.1016 mg/g,and its absorption rate exceeded 44.98 percent;desorpting with 40% ethanol,the desorption rate reached 42.16 percent.The optimal conditions for static and dynamic adsorption of Castanopcis Sclerophylla(Lind1) Schottk flavonoids was that:pH,the sample concentration,ethanol-eluting concentration,and the rate of elution were 3,0.04074mg/ml,40%,2BV/H respectively.3.The study of the anti-bacterial,anti-oxidation of Castanopcis Sclerophylla (Lind1) Schottk flavonoidsCastanopcis Sclerophylla(Lind1) Schottk flavonoids can eliminate Hydroxyl, superoxide radical,DPPH free radical conspicuously.The order of the effects on the removal of free radicals was:DPPH free radical>Hydroxyl>Superoxide radical. Castanopcis Sclerophylla(Lind1) Schottk flavonoids had a certain degree of inhibition to E.coli,Bacillus subtilis,Bacillus Proteus,Staphylococcus aureus,and the capacity to suppression was:Staphylococcus aureus>Bacillus Proteus>E.coli>Bacillus subtilis,yet it hardly inhibited black mold and blue mold.The MIC of Castanopcis Sclerophylla(Lind1) Schottk flavonoids to E.coli is 12.5mg/mL,the MBC of Castanopcis Sclerophylla(Lind1) Schottk flavonoids to E.coli is 25mg/mL. The MIC of Castanopcis Sclerophylla(Lind1) Schottk flavonoids to Bacillus subtilis is 25mg/mL,the MBC of sclerophylla total flavonoids to Bacillus subtilis is 50mg/mL.The MIC of Castanopcis Sclerophylla(Lind1) Schottk flavonoids to Bacillus Proteus is 6.25mg/mL,the MBC of sclerophylla total flavonoids to Bacillus Proteus is 12.5mg/mL.The MIC of Castanopcis Sclerophylla(Lind1) Schottk flavonoids to Bacillus subtilis is 6.25mg/mL,the MBC of Castanopcis Sclerophylla (Lind1) Schottk flavonoids to Bacillus subtilis is 6.25mg/mL.4.The initial study of Castanopcis Sclerophylla(Lind1) Schottk starch preparation and its physical propertiesThe three types of starch paste all were pseudoplastic fluid with shear-thinning property comparing to corn starch and sweet potato starch.By adding 5%NaCl or 10%sugar,the starch paste could improve the transparency.The structure,the rate of frozen-thawed,the mean particle size,solubility and the stability of frozen-thawed of Castanopcis Sclerophylla(Lind1) Schottk starch is similar to corn starch.The hygroscopicity of Castanopcis Sclerophylla(Lind1) Schottk starch is stronger than coen starch and sweet potato starch,but the transparence is lower.The mean particle size of sweet potato starch is biggest of the three types,but the Castanopcis Sclerophylla(Lind1) Schottk starch is the smallest which is easy to make membrane.5.The experimental studies has established the Castanopcis Sclerophylla(Lind1) Schottk starch rheological model was:τ=13.39×γ0.3015,τ=1.9844×γ0.579,τ=13.785×γ0.5559.The study reveals that the ability of anti-cutting of Castanopcis Sclerophylla(Lind1) Schottk starch is stronger than corn starch,but is lower than sweet potato starch.The fluidity of Castanopcis Sclerophylla(Lind1) Schottk starch paste is not better than corn starch,but is better than sweet potato starch.The three types all were non-Newtonian fluid models.6.The hardness of three pieces of starch:the hardness of Castanopcis Selerophylla starch was 45.2g,the hardness of corn starch was 37g,the hardness of sweet potato starch was 69.9g,7.The study of the technology of extracting Castanopcis Sclerophylla(Lind1) Schottk pectinThe extraction factors of Castanopcis Sclerophylla(Lind1) Schottk pectin are: Solid-liquid ratio 1:30,extraction temperature 70℃,extraction time 3h,and pH 2.5. The order of extract factor is pH>solid-liquid ratio>tempreture>time.8.The study the physical properties of Castanopcis Sclerophylla(Lind1) Schottk pectinThe results reveal that galacturonic acid content≥70%,ash content≤5%,Heavy Metal(Pb)≤15mg/kg,As≤2mg/kg,Pb≤5mg/kg,total bacterial count≤100/g, E.coli≤3/100g pathogenic bacteria were found.All the results has met the requirements of the national standard.
