Kenaf, annual herb of Malvaceae Hibscus, is important bast fiber plant. The genetic diversity is abundant. Studies on relationship of kenaf germplasm are important for variety discrimination and widening hereditary basis. Relationship in 38 accessions of kenaf germplasm was assessed using morphology, cytology and ISSR marker.The results are followed:(1) Morphologic investigation results showed that, 38 kenaf varieties had remarkable differences in the shape of bract top,leaf shape,stem color and branch habit. Clustering analysis showed that the materials were divied into six groups at the level of D=0.69.(2)The chromosome karyotypic analysis results showed that the karyotypes of 14 accessions can be described as 2n=36, the karyotypic asymmetry was 55%-60.58%,belonging to symmetrical karyotype. The karyotypic variation ranged within small extension. Karyotypic clustering analysis showed that the materials were divided into three groups.(3)Fiftheen primers were selected from 70 ISSR primers, and 117 DNA fragments were amplified from 38 samples. Of which, the average number of DNA bands was 7.8 per primer. The clustering analysis based on ISSR markers showed that the 38 kenaf accessions could be classified into three groups, at the level of D=0.56.the first group included 19 cultivated species, the differences of genetypes were small,and their genetic basis was comparatively narrow ;the second group included 5 relatives and 7 wild species,;the third group included 1 wild species and 5 relatives.This indicated that there were higher genetic diversity within kenaf cultivated species,wild species and relatives.(4) The results showed that the three methods were consistent basically with comparing classified results, the three methods had respective merits and shortages, ISSR molecule maker was a much effective taxonomic research method.
