Chili pepper is one of the most famous types of dry pepper. Virus can serious infect its production and quality. To research the harm of virus, anti-virus mechanism and the molecular identification methods of main viruses, this research use nine varieties or materials of chili pepper for experiment, testing several main characters. Four varieties or materials of pepper were used for experiment of being inoculated with TMV indoor, testing physiological targets in pepper. The POD isoenzyme patterns in leaves and roots were analyzed, and the virus infected pepper were detected by means of molecular identification methods. The main results were showed as follows:1. Research of chili pepper main character after infected by virus: The results indicated that fruit number, length, weight, seed number and VC content were affected significantly by virus.2. Research of 4 defending-physiological targets in pepper inoculated with TMV: Four varieties or materials of pepper were used for experiment of being inoculated with TMV indoor. By means of measure MDA content , dynamic change of POD, PAL and PPO activities, the results showed that MDA content, POD are higher than CK (non-inoculated). POD, PAL and PPO activities average increased multiple are consistent with resistance, and the average increased multiple of PPO activity is contrary to resistance. For the nine chili pepper materials outdoors, the MDA content and POD activity in the plants which were infected by virus are higher than the healthy plants.3. Relationship between isoenzyme and resistance: Colors of isoenzyme pattern of POD in pepper leaves and roots in inoculated plants were deeper than that in non-inoculated ones, the band of POD were wider in inoculated plants than in non-inoculated. There were 1~2 bands of peroxidase isoenzyme in resistant varieties more than those in susceptible ones.4. Use molecular identification methods to detect TMV and CMV: Based on the coat protein (CP) gene sequence of tobacco mosaic virus (TMV) and cucumber mosaic virus (CMV), 420bp and 780bp fragments were amplified individually by reverse transcription polymerase chain reaction (RT-PCR) with the specific primers.
