The history of XinJiang donkey is very long can ascend to 3rd century, XinJiang is one of main donkey producing area of home donkey, and the number of donkey is large, according to the statistics of 1991, there were 1 million donkey in XinJiang, accounted for 45% of total nantional gross.XinJiang donkey belong to diminutive type in donkey tribes. Originated from, Asian wild donkey came from long time artificial tameness. XinJiang donkey is a good local donkey resource to china with high adaptability, can endurated coarse feedstuff,good reproduction , mature early,growing quickly and others good character.XinJiang donkey mainly distributed at southern area of Tianshan Mountain, In KaShi, HeTian, KuChe,TuLuFan, there are more.Due to the difference of geographic enviroment and ecologic condition, and long time artificial tameness, XinJiang donkey became different populations with different character in these 3 area. In 20th century 90s, Guanzhong donkey had been imparted for hybridization, but at the same time threatened the resource of XinJiang donkey. So, rationally vtilization and protecting peculiar resource of XinJiang donkey improving home donkey`s breeding and production level.cultivating good breed, become a important base to continued development of donkey feeding of XinJiang area.This study was to knowing the genetic varity, define the breeding character of different groups of donkey, provided scientific gist to proteotion and rational utilization for local breed. In this experiment, utilized microsatellite mark technology to study 8 microsatellite loci of 3 local groups of XinJiang donkey, and counted the number of alleles, polymorphism information content , heterozygosity(h) and genetic distances, at the same time made dendrograms.Then analyzed the correlation between different genotypes of microsatelites and physical character. The result as follow:1. In 3 local groups of XinJiang donkey 8 microsatellites loci all showed polymorphisms, in all 74 alleles were observed. Among them, VHL 20 have most allesles,for 11,HTG15 have for 8,The average number of alleles of each microsatellite locus was 9.3.2. Counted the heterozygosity, polymorplism information content and effective number of alleles, and compared genetic differences. The population heterozygosity of 3 local population of donkey ranged from 0.769 to 0.827, the average heterozygosity was 0.789.The polymorphlism information content ranged from 0.655 to 0.867, the number effective alleles aranged from 3.328 to 6.960.3. According to allele frequency, counted Nei genetic distance between Tulufan donkey, Kashi donkey, Hetian donkey, Guanzhong donkey, Dezhou donkey,Jinnan donkey and Biyang donkey(data of Guanzhong donkey, Dezhou donkey,Jinnan donkey and Biyang donkey were cited from reference [48] ). The result showed: genetic distance between Hetian donkey and Dezhou donkey was largest genetic distance ,between Jinnan donkey and Biyang donkey was lowest. Arranged from 0.3365 to 0.9100.4. According Nei`s genetic distance, utilized microware of phylip and treeview , constracted NJ and UPGMA dendrograms. Clustered results: demonstrated 7 donkey populations were clustered into 2 groups, the first group included Guanzhong donkey and Dezhou donkey, the second group included Hetian Donkey, Kashi donkey, Tulufan donkey, all are diminutive type of donkey.5. Utilized SPASS 13.0 microware to analyzed the correlation between genotypes and physical character of hetian donkey. The result showed: there was significantly negative correlation between 190/190 genetype of COR071 microsatellite and hight, bust length.
