| Using 10 microsatellite markers, genetic diversity in ten Chinese domestic donkey breeds(Guanzhong, Dezhou, Jiami, Xiji, Gunsha, Beiyang, Qingyang, Taihang, Xinjiang and Menggu)was estimated. The polymorphism information content, expected heterozygosity and effective number of alleles were calculated, and the Neighbor-Joining tree based on Nei's standard genetic distance was got. The results showed that:1. Six of ten microsatellite loci (HTG7, HTG10, AHT4, HTG6, HMS6, HMS3) were highly polymorphic (PIC>0.5). The HMS7 locus was medium polymorphic (PIC<0.5). The last three loci (HMS1, HTG4, HMS5) were single polymorphic.2. The mean PIC, H, E of seven polymorphic loci in ten donkey breeds were 0.7677, 0.7443, 7.2425. It showed that the Chinese domestic donkey breeds were highly polymorphic and the heterozygosity is relatively high. The seven loci were effective genetic marker to analyze genetic diversity and the dendrogram relationship betweeen different donkey breeds.3. The dendrogram described the genetic relationships between different donkey breeds, which corresponded to their geographic distribution. There were the closest genetic distance among Xinjing, Qingyang, Xiji and Guanzhong donkey breeds. Additionally, Menggu and Dezhou donkey breeds were sorted into one category, which proved more introgression. |
