Study on culture in vitro, in vitro flower and internal variation of physiological and biochemical during flower bud differentiation for Rosa Chinensis minima through tissue culture, can build up Rosa Chinensis minima system of intermediate propagation, decrease the juvenile phases from a vegetative growth to a reproductive growth stage, inquiry into flower bud differentiation regulation is significance not only in theory but also in application.The objective of this research is to study the effects of differ culture medium , 6-BA, NAA, AgNO3, pH and closure film during the induction of lateral bud of Rose Chinensis minima. The results of our research show that germination rate is lower when using B5 and MS culture medium, highest in using 1/2MS. The buds which use 1/2MS culture medium grows stronger, and become formations, the 1/2MS is favorable to intermediate propagation for Rose Chinensis minima. The contrast of differ 6-BA and NAA concentration is very remarkable on the induction rate of lateral bud, also the effects of differ 6-BA and NAA concentration go together with ratio combination are very difference .when 6-BA and NAA all are combined in level of 0.5mg/L, the induction rate of lateral bud is highest at 84.8%, the bud growth is best, higher and orderliness. The effect of differ pH of culture medium is very remarkable on the induction of lateral bud of Rose Chinensis minima, when the culture medium is abnormally high acid or alkali, it will influence the germination rate and go against the growth of bud. As its pH is 5.8, the induction rate is highest, bud grow best and grow many leafs. The effect of differ AgNO3 concentration on the induction of lateral bud of Rose Chinensis minima is significant. When the AgNO3 concentration is 5.0mg/L, the bud growth is consistently more even, certainly prosperous and advantageous to bearing numerous leaves. Through using closure film, the effect on the induction of lateral bud of Rose Chinensis minima also is significance, results shows that closure film can make bud grow rapidly and become a plant. The mostly objective of this research is to study the effects rooting, callus, N/P, sucrose, 2-iP and ABA concentrations on flowering in vitro Rose Chinensis minima. The results of our research show that the plant takes root to repress flower bud inducement obviously, whose floral bud induction was only 1.1%, it is not very impact of floral bud induction whether differentiate callus or not. The results showed that the floral bud induction rate was obviously promoted when we adjusted the N of ammonium nitrate and P content, one of N/P ratio is 1N and 5P (phosphor concentration) can make highest rate of floral bud induction. N and P ratio of contention influence the floral color depth, and the normal growth of plant and leaf. Within limits, the sucrose content of culture medium correlated positively with induct rate of floral bud, but to exorbitant sucrose content will restrain the ratio of floral bud induction seriously. That floral bud induction is highest when the sucrose content is 55mg/L. in addition, the color of flower correlated positively with the sucrose content of culture medium. Effects of differ concentration 2-iP or ABA on floral bud induction of Rosa Chinensis minima were not obvious. The optimal medium that comprehensive various agents which have higher rate of flower buds induction on Rosa Chinensis minima, had the floral bud induction increased greatly reaching 34.4% on the 42 day. The time of comprehensive treatment group on floral bud induction is shorter, higher in induction rate but worse in plant nourishment. We observed some new buds, young leaves and new leaves express for the lilac or the pallor, the color of in vitro flower is shallower than in vivo.In addition, this paper still study on the variation and correlations of physiological and biochemical of Rosa Chinensis minima during the flower bud induction. The results showed that the contention of soluble proteins, soluble sugar, nitrogen and phosphor were higher in control group which floral bud induction is high than in treatment group which floral bud induction is low. Correlations for analysis showed that, the rate of floral bud induction and the contention of soluble proteins, the contention of soluble proteins and nitrogen, the contention of soluble sugar and nitrogen all were strongly positive correlation. The contention of phosphor and the rate of floral bud induction was a negative correlation, but no correlation with nitrogen.
