Disease investigations were conducted in Heba pharmaceutical station, Shankou village, Shiguan village in Shibing Station and so on. Six diseases of Radix Aconiti Kusnezoffii as sclerotium blight, powdery mildew, rust, Pythium disease, leaf spot and Brown patch were recorded. Bubble spots mosaic was caused possibly by birus.The biological characteristics of Sclerotium rolfii were studied. The results showed that amylum was optimum C source and CaNO3 optimum N source for its growth and sclerotia produce; temperatures ranged from 30°C to 35°C were optimum for sclerotium germination. While 30°C was optimum for hypha growth and sclerotia produce. Optimum pH for hypha growth was 5.0 to 7.0 and 3.0 to 6.0 for sclerotium germination. Complete illumination condition promoted hypha growth and sclerotia produce. High RH was beneficial to sclerotium germinationd.The study also indicated that the fungicides, such as 12.5%Diniconazole, gave strong fungistatic action against Sclerotium rolfsii in vitro, with EC500.13μg/ml, and the diameter of inhibition zone 20.6mm.Diniconazole has been determined of the physiological target to Sclerotium rolfii, After dealing with Diniconazole, the content of soluble protein is higher than the comparison. The content of reducing sugar suddenly dropped before 6h dealing, it achieved the lowest at 6h, and it was up to the level of comparison. The content of chitinase isozyme and chitin sugar were higher than comparison. The Diniconazole to the mycelia of Sclerotium rolfsii, discovered appeared the phenomenon of the mycelia cell vesiculated and melting.Investigate and study 5. rolfsii in Lab and field, and the results show which temperature and humidity are the primary effect factors to the sclerotium bligh germation and growth. While no significant efficacy to control sclerotium blight in field. At the time, it provided some theories reference materials for comprehensive control of diseases to screening of biocontrol agent.
