Pepper (Capsicum annuum L.) is an important vegetable, and there are many kinds of carbohydrate, protein, and vitamin in fruit which have high nutritive and health care value. Pepper varieties or cultivar contribute greatly to the efficiency of pepper production,and great attention had been put to the genetic improvement of pepper wordwidly in recently years,especially the application of modern biotechnology in pepper breeding. In this study, In virto and anther culture system of pepper were constructed, and a Agrobacterium-mediated genetic transformation was also constructed with the above In virto culture system, the results were as followings:(1) Pepper seeds stored less than 1 year was chosen for seeding and the derived explant had a high efficiency of in vitro culture; (2) The most suitable explant for in vitro culture was cotyledon of pepper seedling that was 12-16 days old; (3) Multiple buds regenerated directly from cotyledon, a balance of BA and IAA was favorable for multiple buds induction, but high concentration of BA was not favorable to elongation,and addiation of a certain concentration of AgNO3 could help the induction of multiple buds.The most suitable medium for induction culture in our experiment was MS + BA5.0 mg·L-1 + IAA0.1 mg·L-1 + AgNO3 4 mg·L-1 ; (4) There was a significant difference in the induction rate of multiple buds among different genotypes; (5) 1 mg·L-1 GA3 was favorable to the elongation of multiple buds,but higher concentration of GA3 resuilted in more macrocephalic buds, which was not able to be reversed; (6) The best culture medium in our rooting experiment was 1/2MS + NAA 0.1 mg·L-1 + IBA 0.5 mg·L-1 ; (7) The resulte of our genetic transformation experiment showed that when the concentration of carbenicillin was 500 mg·L-1 ,the growth of Agrobacterium could be inhibited, and the mortal concentration of hygromycin to pepper tissue was 80 mg·L-1 ; (8) The best system of Agrobacterium-mediated genetic transformation was 6 day pre-culture of the explants, the OD of the bacterium cell is 0.6, 7 minutes of infection and 3~4 d of coculture; (9) There was a significant difference in pepper anther culture among different genotypes; (10) The phase of microspore development and growth condition were two important factors of pepper anther culture, the anther with high vig or was appropriate and was more suitable for the anther culture; (11) Pretreatment of the anther with low temperature resulted in a lot of dead anther, and the induction effect was lower than high or normal temperature pretreatment; (12) The best culture medium of pepper anther culture was MS + IAA 0.1 mg·L-1 + BA 2 mg·L-1 ; (13) The experiment obtained several anther plants,which was further identified as haploid plants by the modality.
