China was rich in germplasm resources of tea. The genetic diversity and relationship of fifty-one varieties of tea plants in China was studied based on ISSR and TRAP molecular markers. The results were as follows:1. Based on the previous research, according to traits of tea, different parts of tea that were use to extract DNA was compared. A simple rapid and accurate method about extraction and purification of tea genetic DNA were modified from the former method.2. 12 ISSR primers were selected from 40 primers, and 62 bands were amplified by 12 primers, of which 53 was polymorphic. The percentage of polymorphism was 85.5%3. 72 bands were amplified by 18 primer combinations, of which 56 were polymorphic. The average polymorphism was 77.78% and each primer combination amplified 4 bands averagely. The bands were averaged from 100bp to 800bp. The highest polymorphism was 100% and the lowest was 33%.4 according to the ISSR and TRAP results, their dissimilarity coefficient was studied. Cluster analysis dendrogram of tea cultivars was made.
