Study On The Technique Of Chicken Semen Freezing

The test studied dilution, cryoprotectant, equilibrium time and thaw temperature on chicken systemly. Investigated the differences of frost resistance individualy .Meanwhile studied the influence about radix astragali, angelica and radix codonopsitis on chicken semen quality.The purpose is to set up chicken semen cryopreservation technique system. To supply technique support for chicken genetic resources protection, new species breeding and manufacture.The result showed that:Semen diluent influence cryopreservation:Choose 6 semen diluent to filter appropriate chicken semen cryopreservation diluent. The diluent contain magnesium chloride, potassium citrate, sodium glutamate, dipotassium hydrogen phosphate, monopotassium phosphate and fructose, semen vitality is hightest after frozen.The average value is 0.52,highter than other 5 diluent significantly (P﹤0.01).So chose this diluent as semen cryopreservation diluent on chicken.Definite cryoprotectant, equilibrium time and thaw temperature:On the base of test one,research freeze sheltering effect about glycerin, DMSO ,DMA and DMF, each with 4 consistency.The result showed that 10%DMSO sheltering effect is the best, the next is 4%DMA,semen vitality are 0.58 and 0.55 after frozen.Haven no difference (P﹥0.05). Based on the result ,chose 10%DMSO as cryoprotectant, compare the influence on semen vitality after frozen at different equilibrium time and thaw temperature. Equilibrium time is 30mins and thaw temperature is 40℃,semen vitality is best after frozen.Chicken semen frost resistance among chickens:chose 10 rooster to research the difference of semen frost resistance individually.The result showed semen vitality have significant difference after frozen(P﹤0.01).Number one rooster's semen vitality is hightest after frozen ,is 0.56.The result showed chicken semen has significant difference on frost resistance.The influence about Chinese medicinal herb additive on chicken semen quality: experimental group compared with control group,except semen density,semen quantity,semen vitality and malformation rate all have significant difference.
