| The experiment used the medium-speed Haiyang Yellow Chicken three-line matching family cock as the material,a single-factor complete randomized trial design method was used to compare the differences in semen quality among 30 individual cocks in three parental groups,reveal the semen quality characteristics of the three parental groups and provide basic data for the subsequent improvement of reproductive performance of the cock,at the same time,the semen of the FF group cock was used as the test material;Using the semen dilution developed by Jinghai Group to study the effects of different dilution factors and different placement time on the quality of semen,find the optimal dilution ratio,and propose the preservation time under different dilution ratios to provide reference for artificial insemination of chickens;Finally,the effects of different storage time on the quality of semen after high temperature sterilization and double-antibody dilution were studied to solve the problem of the shelf life of semen dilution.The research of the thesis focuses on improving the quality of the cock's semen,improving the breeding value of the cock,ensuring the fertilization rate,thereby reducing the breeding amount of the cock and saving the cost of cock breeding.Below are major research results:1.There is a certain difference in semen quality between the three lines of Haiyang Yellow Chicken Parents.The semen volume,sperm density,viability,plasma membrane integrity and effective sperm count of FF cock semen are significantly higher than JJ and BB.Cock(P<0.01);FF cock semen deformity rate was significantly lower than BB cock(P<0.01),but there was no significant difference with JJ cock(P>0.05).2.Effect of different dilution ratios of specific dilutions on semen qualityThe semen activity diluted 1:2 was significantly higher than the original semen and 1:1 multiple dilution of semen after 2 hours of in vitro storage(P<0.01).The 1:2 multiple dilution of semen was significantly lower than the spermatozoa.1:1 multiple dilution and original semen(P<0.01),different dilution factors had no significant effect on the malformation rate of semen(P>0.05);after 2 hours of storage in vitro,the 1:2 dilution of semen was higher than 1:1 dilution and original semen,dead sperm rate and deformity rate are significantly lower than 1:1 dilution and original semen,1:1 multiple dilution of semen is the worst,1:2-fold dilution is best.3.Effect of different dilutions on semen quality during storage at different timesThe semen activity of the self-dispensing dilutions at 0.5 and 1.5 hours was significantly higher than that of the basic dilution and BPSE dilution(P<0.05).The semen malformation rate of the BPSE dilution was significantly greater than the basic dilution at 0.5 and 1.5 hours.Self-mixed dilutions(P<0.05);but the effects of the three dilutions on sperm death rate were not significantly different(P>0.05).4.Effect of different treatment dilutions on semen quality at different timesAfter the high temperature sterilization and double-antibody treatment,the dilutions were stored for 3 days and 6 days,respectively,and the semen quality at these two time points was examined.The results showed that there was no significant difference between the two treatments at different storage time points,but the dilution effect of the high temperature sterilization treatment was better than that of the double treatment treatment. |
