The objective of this study was to map the gene determing self-compatibility (SC) in the F2 population derived from a cross between Eruca sativa. Mill.self-compatible line ( WuYun 7511 ) and self-incompatible line ( Bolish ).In addition,the common SSR primers were selected between Eruca sativa. Mill. and Brassica rape varieties. The results are as follow:1. In the BC1F1 and F2 population appears self-compatible plants and self- incompatible plants. Chi-squared (χ2) tests were applied to the inheritance data in the F2 population. The F2 population self-compatibility separation conforms to the single gene heredity.It indicated that Eruca sativa. Mill. self-compatibility gene was possibly controlled by single gene .2. The most suitable SSR system for the research was set up.The SSR reaction mixture was 20ul volume: DNA (0.5ul) +Mg2+ (2.0ul) +Tag (0.5ul) +dNTPs (2.0ul) + Primer (1.0ul). The suitable reaction procedure was: 94℃,5min;35 cycles: 94℃, 50s, 55℃, 50s, 72℃, 1min; the last 72℃,10min.3. Through the Bulked Segregant Analysis (BSA) method, three out of 29 primers had amplified polymorphic fragment among self-incompatible and self- compatible pool.But only Na10- F06 had polymorphic fragment in F2 population。Using Na10-F06 to detect the genetic distance between the marker and the gene determing self-compatibility by means of the Kosambi mapping function.As a result,it proved to be 10.13cM.4. Twenty-nine pairs of SSR primer were amplified between Eruca sativa. Mill and Brassica rapa.The results indicated: three primers showed polymorphic in the Eruca sativa. Mill. self-compatible plants and self-incompatible plants,but nine pairs of the primers had multi-state properties in the eight Brassica rape; the fragment size and the fragment number amplified by the same primer between Eruca sativa. Mill. and Brassica rapa was different.This study makes the important progress on SSR marker of gene determing self-compatibility in Eruca sativa Mill.,and obtained a likaged SSR marker. The selection of common SSR peimers in the different genus have a important effect to the SSR marker of different species.
