In order to explore the genes of differential display in the course of selecting follicular cells, egg-laying Yangzhou goose was chosen as the research material in this paper.By using differential display reverse transcription PCR that piloted by combinations of three anchor primers and twenty-three random primers, eighteen different expressed fragments were obtained, which were separated by polyacrylamide gel electrophoresis products of different follicular cDNA. After Re-PCR, electrophoresis and purification, ten different expressed fragments were got. After comparing the results of Re-PCR and that of the first PCR and then dot-blot detection, there were still 7 genuine differential display fragments. Five ESTs sequences were obtained by clone and sequencing by Shanghai Jierui Biological Engineering Technology & Services Co. Ltd. They were short in length and feeble in brightness. The reason might be the low abundance of differential display genes.After getting rid of vector sequences, the five ESTs were submitted to Blast. Among these 5 fragments, two of them were homology with known gene, they were, EST_YZGF01 homology with human MAT IIβsubunit and the homology 95%, and EST_YZGF02 homology with chicken mKIAA0840 protein and the homology 94%, respectively; the three others were not homology with any known gene or sequence suggesting that they might be new genes. The increasing expression level of the smallest preovulatory follicle(SPF) MAT IIβsubunit may be beneficial to follicle selection by providing more substrates to synthesize E2 or toning up expression level of anti-apoptosis genes. FBXL-7 might be a part of SCF complex, and involved in follicle selection through ubiqiutin-induced degradation of transcription activation factor or inhibiting factor.
