The terminal and axillary buds of 'xueqing', 'lvbaoshi' and European pearrootstock 'BA29', and leaves of 'BA29' were used as explants in this study of organculture to establish a rapid technique system of micropropagation. Orthogoal designand analysis of variance and multiple comparisons with DPS analysis software wereused in this experiment. The experiment was mainly concentrated on initiation,multiplication stage of 'xueqing', 'lvbaoshi' and 'BA29', together with rooting,regeneration stage of 'BA29'. The results were as follows:1. Using four different sterilization time, the most effective sterilization (HgCl2 0.1%)time for "xueqing' was 8 minutes . But the best sterilization time for 'lvbaoshi' and'BA29' was 5 minutes each.2. Using three different size explants, the size of 0.5 cm long stem segment was thebest for 'xueqing' and 'lvbaoshi'.3. Using of 0.1% HgCl2 was the best sterilization chemical for explants compared to0.1% HgCl2 followed by 75% spirit for 'xueqing' and 'BA29'. 'Lvbaoshi' was notsensitive to the methods.4. Picking 'lvbaoshi' explants in April, May, June, July, April was the best time forpicking the explants of 'lvbaoshi'and had the best sprouting effect.Putting theexplants in the refrigerator, using of medium with 100mg/L VC, using with 2g/L PVP,medium with 100mg/L VC and 2g/L PVP were more effective for reducing browningrate of 'lvbaoshi'.5. The initial medium of 'xueqing', 'lvbaoshi' and 'BA29' were MS+BA1.0mg/L+IBA0.1 mg/L.6. The multiplication medium of 'xueqing' was MS+BA2.0mg/L+IBA0.2mg/L,'lvbaoshi' was 1/2MS+BA 1.0mg/L+IBA0.1mg/L, and 'BA29' was MS+BA 1.0 mg/L+IBA0.1mg/L.7. With the increasing dose of GA3 (0mg/L,0.5mg/L,1.0mg/L,1.5mg/L),multiplication of the stems of 'xueqing' and 'lvbaoshi' were induced. Butmultiplication of stems of 'BA29' was decreased when the concentration of GA3 wasmore than 1.0mg/L.8. The rooting medium of 'BA29' was 1/2MS+IBA 1.0 mg/L+ NAA 0.3mg/L。'BA29'rooted better after 5 days in the dark. 9. The leaves of 'BA29' were cultured for 16, 20, 24 days in the dark. With the darktime increased, the regeneration frequency of 20 and 24 dark days was increased. Theregeneration frequency and buds number per leaf of 'BA29' leaves were higher whenabaxial side was placed downward onto medium than upward. The leaf regenerationmedium of 'BA29' was MS+BA 5mg/L+IBA 0.5mg/L. |
