Effects of medium composition, vessel types and air exchange of vessel onrapid propagation of virus-free sweet potato in vitro were studied. Also, the appropriate wayof producing seed tubers and optimizing extension system with plantlet of virus-free in sweetpotato were discussed. The results showed that 30 mM of total nitrogen, 1:1 of NH4+ andNO3- ratio, 50 g/L of edible sugar in MS medium, polycarbonate (PC) vessel and 1.0 cm ofair exchange diameter were suitable for proliferation and growth of plantlet in virus-free ofsweet potato.Procedure of producing seed tubers in virus-free included breeding of breeder's seed,basic seed, and high quality seed. The breeding of breeder's seed must use virus-free plantletsin the house having insect-proof net, and the soil must not have pathogenic bacteria/fungi.The production and environment conditions of basic and high quality seed breeding weresimilar to generic sweet potato. Furthermore, virus-free seed of sweet potato should not beused continuously for two years in the culture. The extension system of virus-free plantletshould be consummated, the standardize, market should be established. In order to enhancingthe quality of seed tubers, scaling-up propagation, and increasing yield, the virus check needbe enhanced.
