The neuropeptide is the bioactive polypeptide which distributes mainly in the nervous tissue, the immunity, the cardiovascular, thereproduction or other tissues, recently. They have their influence on various immunocyte by means of neuroendocrine which participte immunomodulatory and promote immunity function. When the ostrich were toartificially tamed from the wild condition, specially introduced from original habitat Africa to our country, its survival environment, the life habit and the food had all changed a lot. So it unavoidably appeared such as the weaken resistance, the reduced productivity or other adverse reactions.The experiment research the distribution of neuropeptide Y (NPY) and substance P (SP) in the main immune organ (spleen and thymus) of ostrich during difference age stage with the immunity histochemistry SP method, Which provides the theory basis for the research of occurrence and therapy mechanism of disease as well as scientific raising of the ostrich. The results are as follows:1. In the thymus, the NPY nerve fiber distributed mainly in peripheral blood vessels,interlobular septum and the junctional zone between cortex and medulla, capsule which distributed a little. The vessel wall of capillary in the junctional zone between cortex and medulla, thymocyte, paracortical zone contained NPY nerve fiber too. NPY immunopositive cells are circular or oval which moderately dyed and showed light blue or the blue color, the minority slice showed the deep blue. Cytoplasm dyed homogeneously as well as the cytomembrane. NPY immunopositive cells are inhomogeneous distributed in the arterial canal and interlobular septum of the ostrich when the young . Ater the first month, the NPY nerve fiber had appeared. immunopositive cells increased continuously in the adolescence. The number of NPY immunopositive cells are most and tended to be stable in sexual maturity.2. In the spleen, the NPY nerve fiber circleround in splenic artery, splenic vein, the adventitia and tunica media of trabecular artery and trabecular vein. The NPY nerve fiber distributed abundantly in the lymphoid theca which distributed the artery of white pulp and surround the central artery of acinus lienis. The nerve fiber distributed the smooth muscle layer of media blood vessel, mainly by circumduction, during alternates little longitudinal courser the fiber, mutually interweaves the net. NPYnerve fiber were found in capsule, trabeculum, medulla, splenic cord and marginal zone. NPY immunopositive cells were circular or oval, and dyed blue which located a lot in white pulp and marginal zone, but never found in red pulp. In the young and adolescence period, the number of NPY immunopositive cells in the spleen of ostrich increased along with the age, and reached most when the sexual maturation period. But this kind of growth tendency is not obvious.3. In the thymus, SP nerve fiber showed the striation or the string of beads shape, which were rich in capsule and interlobular septum, walked along the blood vessel mainly. Some fiber also had entered into the junctional zone between cortex and medulla of parenchyma, medulla and lymphoid tissue of cortex along with connectivum of lobule. It also distributed in the thymocyte of cortex and master cell. The SP immunopositive cells in the thymus assumes deep blue either the blue-black circular or oval, mostly. Endochylema and cell membrane were stained together and stained deeper from central cells to cell membrane. All the cell membrane were strongly stained than cytoplasm. The ostrich thymus had a few SP immune response masculine cells, the earlier period has not contained the SP nerve fiberdistribution, after first month gradually appears. The adolescence, the masculine cell number obvious are many to the young age time. The natural mature period, the masculine cell number gradually decreases,the masculine nerve fiber also gradually decreases.4. In the spleen, SP immunity masculine nerve fiber distribution showed a typical linear pattern, with non-obvious varicosity. The nerve fiber walks along the branch of splenic artery mainly, especially rich around the central artery, longitudinal courser is main modality. The lymphatic tissue in white pulp and red pulp also has a little SP masculine textile fiber distribution, and has the contact with the lymphocyte and the spleen. The SP immune response masculine cell assumes the circular or ellipse, dyes must be shallower than the thymocite, assumes the blue color or the deep blue. In the all physiological time, the number of SP immuneresponse masculine cell in the ostrich spleen and the ostrich age change presents the correlation, but the relevance is not obviousIn view of the above, we draw such conclusion:1. The immunomodulatory mechanisms of NPY and SP in the ostrich thymus might be related to the adjustment of thymus development.2. Besides adjustment of lymphocyte to release lymph factor, NPY and SP succeed in spleen immunomodulatory by the control of spleen blood current capacity regulation, possibly.
