Agrobacterium-mediated transformation of stilbene synthase gene (STS) from Chinese wild Vitis pseudoreticulata in Vitis vinifera L.'Emerald Seedless'was studied. In addition, previous obtained'Thompson Seedless'hygromycin-resistant lines were assayed by PCR,Southern blot and HPLC. Main results were obtained as follows:1. leaf and petiole of'Emerald Seedless'were used in this study. Different combination of plant growth regulators were studied to induce the adventitious buds. Result shows that the best medium in inducing the adventitious buds was MS+BAP 6.0 mg/L+NAA 0.2 mg/L+sucrose 30 g/L+ agar 0.6%+ CH 200 mg/L + PVP 500 mg/L in our experiment. And the regeneration efficiency was 22% and 13%, respectively.2. Agrobacterium-mediated transformation of leaves and petioles of'Emerald Seedless'was conducted and 7 hygromycin-resistant plantlets were obtained.3. 27 hygromycin-resistant lines obtained in our previous research were assayed by PCR, of which 11 lines were STS-positive. When assayed by Southern blot, 5 of the 9 STS-positive confirmed by PCR were positive. And the STS-positive line STS 7 confirmed by Southern blot was assayed by HPLC. Results show that the resveratrol content in transgenic line STS 7 was 5.5 times higher than non-transformant.
