Chicken-quail distance hybridization existed incompatibility to some extent.Most of the chicken-quail hybrids died in early embryo period.Follow the study of chicken-quail distance hybridization, in order to overcome chicken-quail interspecific cross incompatibility, the certain time when most of chicken-quail hybrids died is needed to determine. For the purpose of development this study,the relation between the distinction of differentially expressed protein of chicken or quail or the hybrids in early embryo period and the death of the most of the chicken-quail hybrids.The experiment investigated chicken (♂) and quail's (♀) early hybridization embryos.Frist,1494 hatching eggs of hybrids was taken at random to investigate the morphology of hybrids embryos,the result is that the certain time when most of chicken-quail hybrids died is 72±6h after eggs was hatched.Second, analyse the distinction of differentially expressed protein of chicken-quail hybrids by SDS-PAGE, the analyse of the distinction of differentially expressed protein of chicken or quail was kept for reference.The result shows that the protein bands of 66,200Da express the different type in early hybridization embryos, while the same type in chicken or quail early embryos.There are thick protein bands of 66,200Da express in early hybridization embryos of 72h after eggs was hatched.Third, Qwpkci1 primers was designed referencing the Wpkci gene exon3(3316~3789bp) in quail's W chromosome, and the gene is special in sexing the quail.The primer amplified the cDNA of the early hybridization embryos and obtained one special bands in female one ,while male one received nothing.β-actin primers was designed referencing Gallus gallus cytoplasmic beta-actin gene. The primer amplified the cDNA of the early hybridization embryos and obtained one special bands both in in female one and in male one. Qwpkci1 primers andβ-actin primers was used in the same system of PCR to sexing the early hybridization embryos whose expressed proteins is diffent by SDS-PAGE.The result shows that the one who has thick protein bands in early hybridization embryos of 72h after eggs was hatched is female,while who has not is male. The overexpression of protein of 66,200Da in early hybridization embryos of 72h after eggs was hatched may be have main infection with the early embryonic development.
