The type and amount of vertebrate muscle fiber are determined by genetic factor,they formed during the embryo or fetal period.But it is an important stage for the metabolism of muscle fiber and the type transforming of contraction at early growth phase of the postnatal.The quantity of muscle fiber is no longer changed after the birth of the animal, the extent and the diameter of muscle fiber is increased for the growth of muscle,but not the proliferation of cells and the increasing of myocyte amount. The development condition is directly determined the production and the quality.So the potential of meat producing and the quality of animal muscle are the key to the early development of muscle. The meat quality,the segregation and combination of correlation gene of chicken-quail hybrids is also existed in the parents.In order to revealing the essential rule of muscle development,beef using and egg using cocks were selected as male parent,quails were used as female parent for cross-fertilizing.Nineteen embryos of muscle tissue for thirteen-day beef type hybrid species and twenty-one embryos at the same period of egg type hybrid species were prepared for SSH(supp ression subt ractive hybridization),these tissues can ensure at least four times for this study.In this research,the beef type hybrid embryos were used as experimental group,while the egg type hybrid embryos were used as control group. For undertaking the forward direct SSH, decrease lib for muscle development is constructed.180 cloning colony were got by blue-white blot, through PCR detecting for the use of Primer 2R and Nested Primer 1 to every clone, recombined clone were selected and identified for the size of insertion element.140 positive clones were selected from the construction of cDNA SSH libraries,except negative clones and multitude strap clones.The size of insertion element mostly distributed from 250bp to 750bp. Random selection for 100 positive clones were selected for sequencing.Based on the homology Analysing for the result of sequencing,4clones of 40 masculine clones are highly homologised to Y-box conjugated protein,FK506 conjugated protein 3, Vimentin and somatic muscles troponin T,respectively.Although there is no report on the gene sequence,the homologies are all higher with the comparison to other cloned sequences and the ESTs of chicken BAC Lib.
