The embryonic development of birds is complex, leading to embryonic death by many factors. There is a peak of death in the early embryonic development of hybrids of chicken-quail,with male and completely sterile identificated in individuals hatched to the of after all. These phenomena can not explained with the reseans of death of embryos in common sense. In this experiment, we observed morphology of hybrids of chicken-quail. Then the embryos of chicken.quail and hybrids of chicken-quail were injected by Formestane and 17β-estradiol to observe the influence of Formestane and 17(3-estradiol in sex differentiation of quail. This experiment was taken with explaining relation between laws of ER gene and so on expression in the embryos of chicken.quail and hybrids of chicken-quail treated by drug before and after and early death of embryo of hybrids of chicken-quail.1.Morphological observation on the development of embryos of hybrids of chicken-quail,quail, chickenAt present, there are many intimate reports on study of embryonic development morphous of chicken and quail.but no reports on hybrids of chicken-quail. We researched hybrids of chicken-quail, quail, chicken by anatomical micro scope and taking pictures from brooding to birth. The results showed that the hybrids of chicken-quail are a little different from chicken and quail on morphological development in the whole. The hybrids of chicken-quail are faster than chicken and slower than quail in the space and there are big differences in the hybrids of chicken-quail,quail,chicken.2. The study of sex reversal of chicken embryo and quail embryo induced with Formestane and 17-estradiolThe embryo sex of pristine poultry breeding is affected by gonadal hormone. The sexuality of normal hatching egg injected 17β-estradiol and arimedex is induced to maleness femina. To authenticate influence of Formestane and 17(3-estradiol on sex differentiation of chicken and quail embryo, according to the mechanism of sex differentiation in poultry,3 days chicken embryo and 2 days quail embryo were injected by 100μg/ml Formestane and 50μg/ml 17β-estradiol to observe the influence of Formestane and 17β-estradiol in sex differentiation of quail. We found that the chicken embryo and quail embryo injected Formestane and 17p-estradiol had sex reversal. At the same time, the hybrids of chicken-quail were treated by the means the same to quails used. But the result displays that death rate is high and there is no one living. On these grounds, we conjecture that disorders of gene expression caused by the medicines injected lead to death of the whole hybrids of chicken-quail.3. Expression analysis of ER,3β-HSD,P450c17and P450arom geneBird embryos at the early sexual differentiation affected by sex hormones, especially estrogen plays a very important role in the sexual differentiation. The body of estrogen is generally produced by the conversion of cholesterol, the transformation process required hydroxy steroid dehydrogenase (mainly have 3β-HSD and P-450c17 gene in poultry) and P450arom gene. In this experiment, we collected ten embryos of the chicken, quail and their intergeneric hybrids at72h,96h,120h,144h,168h and 192h time point, basing the sexing of embryos, using Real-time RT-PCR method to obtain the target gene expression. Hybrids show the trend of expression in advance.ER early expression of the volume also showed their parents have greater differences, while its expression time and their embryos consistent with the time of death basically. Therefore, we conjecture that ER gene take a key role at the chicken and quail embryos intergeneric hybridization study of early death.4. Expression analysis of bcl-2 and p53 geneAt present there is no reports about the expressive pattern of bcl-2 and P53 of each period of early embryos. In this experiment, we collected ten embryos of the chicken, quail and their intergeneric hybrids at72h,96h,120h,144h,168h and 192h time point, basing the sexing of embryos, using Real-time RT-PCR method to obtain the target gene expression. We found that the expressional peak of bcl-2 gene expression of female embryos coincide with its P53/bcl-2 lowest expressional level and to compare chicken and quail, the bcl-2, P53 gene expression of early embryos of hybrids were abnormal at 72h.120h. Therefore, we conjecture that the abnormal expression of bcl-2, P53 gene may take a role at the chicken and quail embryos intergeneric hybridization study of early death.5. Expression analysis of ER gene and so on induced with Formestane and 17-estradiolThe sex hormones take an important role in the early sexual differentiation of bird embryos. We collected embryos of chicken and quail induced with Formestane and 17-estradiol in different periods,using Real-time RT-PCR method to obtain the target gene expression. The results showed that expression of ER gene and so on appears disorders.The condition is consistent with high death rate ofmbryos of chicken and quail induced with Formestane and 17-estradiol.
