Mastitis was always one main reason that affected economic efficiency and rapiddevelopment of dairy goats and cattles. It affected milk yield of dairy goats and cattles. Iteliminated serious dairy goats and cows and resulted in serious economic losses. It also affected thequality of milk and endangered people's health. It have been attracting widespread attention inanimal husbandry and veterinary, food and other fields. Our country and foreign countrysconducted a lot of researchs about the morbidity reasons, pathogenesis, prevention and treatment ofmastitis and produced different theorys. But oxidative damage and antioxidative system of mastitiswere reported very few.The topic studied dairy goats of control group, vaccine group, traditional Chinese medicinegroup, combination group and mastitis group in lactating. The topic detected free radicals in serumand mammary gland, the fluidity of mammary epithelial cells envelope, DNA oxidative damage ofmammary gland, ultramicrostructure of mammary gland, antioxidase, antioxidant agent, antioxi-dative microelement and total antioxidative capability in serum and mammary gland of dairy goats.The topic systemly researched the change of oxidative damage and antioxidative system in serumand mammary gland in dairy goats with mastitis in lactating and investigated oxidative damage andmechanism of mastitis. The results showed as follows:1.The content of active oxygen, NO, MDA and the activity of NOS in serum and mammarygland in dairy goats with mastitis were significantly higher than other groups(p<0.05, p<0.01);Vaccine group were significantly lower than mastitis group(p<0.01); Traditional Chinese medicinegroup were significantly lower than control group(p<0.05, p<0.01); Combination group weresignificantly lower than vaccine group(p<0.05, p<0.01); Combination group were not significantlydifferent with traditional Chinese medicine group(p>0.05). The results showed that free radicalsaccumulated in the body and mammary gland in dairy goats with mastitis. These free radicalsresulted in oxidative damage. Mastitis vaccine and traditional Chinese medicine could inhibitexcessive free radicals and weaken lipid peroxidation of dairy goat.2. The micro-consistency of mammary epithelial cells membrane in dairy goats with mastitiswas significantly higher than other groups(p<0.01); Vaccine group was significantly lower thanmastitis group(p<0.01); Traditional Chinese medicine group was significantly lower than controlgroup(p<0.05, p<0.01); Combination group was significantly lower than vaccine group(p<0.01);Combination group was not significantly different with traditional Chinese medicine group(p>0.05).The results showed that the fluidity of mammary epithelial cell membrane was disordered andaffected normal function of cellular membrane in dairy goats with mastitis. Mastitis vaccine and traditional Chinese medicine could prevent free radicals to damage mammary epithelial cellsmembrane.3. The DNA fragmentation rate(%) of mammary gland in dairy goats with mastitis wassignificantly higher than other groups(p<0.01); Vaccine group was significantly lower than mastitisgroup(p<0.01); Traditional Chinese medicine group was significantly lower than controlgroup(p<0.05, p<0.01); Combination group was significantly lower than vaccine group(p<0.05,p<0.01); Combination group was not significantly different with traditional Chinese medicinegroup(p>0.05). The results showed that DNA of mammary gland suffered serious injury in dairygoats with mastitis. Mastitis vaccine and traditional Chinese medicine could prevent free radicals todamage DNA of mammary gland.4. The activity of antioxidases in serum and mammary gland in dairy goats with mastitis weresignificantly lower than other groups(p<0.05, p<0.01); Vaccine group were significantly higher thanmastitis group(p<0.05, p<0.01); Traditional Chinese medicine group were significantly higher thancontrol group(p<0.05, p<0.01); Combination group were significantly higher than vaccine group(p<0.05, p<0.01); Combination group were not significantly different with traditional Chinesemedicine group(p>0.05). The results showed that the activity of antioxidase decreased in body andmammary gland in dairy goats with mastitis. Mastitis vaccine and traditional Chinese medicinecould enhance the activity of antioxidase of dairy goats.5. The content of vitamin E and vitamin C in serum and mammary gland in dairy goats withmastitis were significantly lower than other groups(p<0.01); Vaccine group were significantlyhigher than mastitis group(p<0.01); Traditional Chinese medicine group were significantly higherthan control group(p<0.05, p<0.01); Combination group were significantly higher than vaccinegroup(p<0.05, p<0.01); Combination group were not significantly different with traditional Chinesemedicine group(p>0.05). The results showed that the decreasing content of antioxidative vitaminsweakened antioxidant capacity in dairy goats with mastitis.6. The content of Se and Zn in serum and mammary gland in dairy goats with mastitis weresignificantly lower than other groups(p<0.05, p<0.01); Vaccine group were significantly higher thanmastitis group(p<0.05, p<0.01); Traditional Chinese medicine group were higher than controlgroup(p>0.05); Combination group were not significantly different with vaccine group andtraditional Chinese medicine group(p>0.05). The results showed that the decreasing content of Seand Zn weakened antioxidant capacity and resistance in dairy goats with mastitis. The content of Cuin serum and mammary gland in dairy goats with mastitis was higher than other groups(p>0.05);Other groups were not significantly different(p>0.05). The mechanism of increasing content of Cuwas still not very clear.7. The total antioxidant capacity(T-AOC) in serum and mammary gland in dairy goats withmastitis was significantly lower than other groups(p<0.01); Vaccine group was significantly higherthan mastiffs group(p<0.01); Traditional Chinese medicine group was significantly higher thancontrol group(p<0.01); Combination group was significantly higher than vaccine group(p<0.05, p<0.01); Combination group was not significantly different with traditional Chinese medicinegroup(p>0.05). The results showed that the total antioxidant capacity decreased in dairy goats withmastitis. Mastitis vaccine and traditional Chinese medicine could enhance the total antioxidantcapacity of dairy goats.8. The ultramicrostructure of mammary gland were injured in dairy goats with mastitis. Theresults showed that free radicals caused oxidative damage of ultramicrostructure in mammary glandby lipid peroxidation. The ultramicrostructure of mammary gland in other groups were normal. Itshowed the enhancement of antioxidant defense system effectively controled over oxidationdamage of mammary gland caused by peroxidatic reaction in vaccine group, traditional Chinesemedicine group and combination group.The study revealed that the imbalance of antioxidant defense system(antioxidant enzymes,antioxidants), the increasing of reactive oxygen and nitrogen free radical and enhanced lipidperoxidation resulted in DNA fragmentation, the decreasing of membrane fluidity and oxidativedamage of mammary tissue in dairy goats with mastitis. Thus mastitis vaccine, immunoenhancer oftraditional Chinese medicine and their combination application could significantly enhance theantioxidant function of antioxidant system and reduce oxidative damage of mammary tissue. Theyguaranteed health and normal function of breast in dairy goats and provided a new way for theprevention and treatment in mastitis of milk animals.
