Red deer is a rare and wild animal in our country, and it is the second class wild animal whichis protected concemfully. Renowned for its production of young pilose antlers and high production,the artificial domestication and breeding of red deer has developed rapidly in recent years. Asianwapiti for this experiment to test our targets, we studied the electroejaculation and semen'scryopreservation. It was the most crucial link of artificial reproduction for the farming system. Theaim of study was to upbuild a set of perfect protocol of artificial reproduction that was feasible tored deer in the practice, so an to give theoretic basis, experience and methods to the deep study ofdeer breeding. In the period form September 2005 to October 2006, the experiments wereconducted at Guanjia deer farm, the Manchu Autonomous County in Liaoning Province, whichincluded the semen collection, freezing of semen and so on. The results were as follows.1. An electroejaculation technique was applied to red deer for semen collection andcharacterization of their seminal traits. Semen electroejaculation were recovered from 68 of 18deers. Ejaculation of deer is 58 times, and the percentage of success is 85.29%. The voltage ofejaculation was 8±1.72V. The volume of semen was 3.29±0.67mL.The motility of sperm was0.82±0.091.The consistency of sperm was 11.16±1.52×10~8/mL.The percentage of abnormal spermwas 13.74±1.836%.The percentage of intact acrosome was 85.49±2.13%.The consistency of mostbreeding deer was dense.2. Lumianbao was a more suitable narcotic than others to electroejaculation. Thepharmaceutical dosage was 0.012mL/kg. The narcotic was effective, safe and indeed, and setted outa smooth recovery using with Luxingbao. The various physiological functions of red deers werewell. After the deers were anesthetic, only three men were able to take electroejaculation.3. In the process of electroejaculation, the result of ejaculation and the quality of semen wasthe best at the same time of the wapiti's genital was the whole erection.4. Different time in the breeding season, semen quality of breeder deers showed a downwadtrend as the continued time. Different times of electroejaculation in breeding season, semen qualityof breeder deers showed a downwad trend as the increasing times.5. Three of the dilutions, the best one was ingredientⅢ(AndroMed) on the ability offertilization. After frozen-thawed, the spermic activity of ingredientⅢhad very significant,difference with ingredientⅠ(Tris+citric acid+glucose) and ingredientⅡ(sucrose). The betterdilution was ingredientⅠ. 6. In four treatments, the best equilibration time was 4 hours. After frozen-thawed, thepercentage of active spermatozoas was not significant difference with it of treatmentⅠ, and wasvery significant difference with it of treatmentⅡ, and was significant difference with it oftreatmentⅠ. Comparing the effect of various thawing temperature, the best was 40℃and settling10 sounds. After frozen-thawed, the percentages of spermatozoa motility was very elevatedremarkably comparing with it of thawing temperature 36℃.
