The test studied on the freezing dilute and freezing method of chicken semen. First, we designed 8 dilutes, then using them freezed chicken semen respectively for six times, according to freezon-thawed semen quality we finally got one better dilute. On the base of NO.1 dilute, we studied the effect of osmotic pressure, cryoprotectant, yolk, caffeine and vitamin E etc. on freezing semen. Finally, the freezing method of chicken semen including the speed of tuning down the tempreture and so on was studied. At last, we got a suitable freezing programme. Bellow are the some of results:1. NO.1 dilute was the best one in this studying, its osmotic pressure was 422mOsm and pH was 6.48. The sperm vitality was 0.72 highter than other 7 diluents significantly (P﹤0.01).2. When the osmotic pressure was 420mOsm, we got the best freezing result. It significantly highter than 330mOsm,360mOsm,390mOsm,400mOsm and 440mOsm (P﹤0.01).3. 6% glycerin was the best cryoprotectant, then was 10%DMSO, finally were 10% DMA and 4% glycol. The result of double cryoprotectant did not have significantly difference.4. When 10% yolk, 5mmol/L caffeine and 15mg/mL vitamin E were added, the freezon-thawed semen quality was improved significantly.5. When chicken semen was freezon, the speed of turning down the tempreture should be controled at 0.55℃/min, and then hold at 4℃for 120min.6. On freezing chicken semen, the pellet frozen semen was better.7. The frozen semen should be thawed dryly, and the number of pellets should be less.8. Using a higher tempreture to thaw the freezon semen was better than a low one.9. If using centrifuge to put the glycerin out from dilute, the speed should be controlled at 800rpm.
