Manhukang oral liquid was consist of seven Chinese traditional herbs, including baicalin,stemona and honeysuckble and so on. It possessed the effect of moistening lung, arresting cough,heat-clearing and detoxicating, It was mainly used to prevent and cure Mycoplasma gallisepticum,which can cause chronic respiratory passage disease in chicken, and the respiratory passagediseases originated for other reasons, This oral liquid possessd two characters of improvingimmunological functions and lower resistance to bacteria. For the further purpose to explor and usethe oral liquor, according to the research of neo-veterinary drugs, a systematic research was carriedout in this paper on preparing process, quality control standard and preliminary stability of thispreparation.In the preparing process, for the major component--baicalin in this preparation, decoctionfrequency, quantity of water, content of tween-80, incubation time of acid praecipitatum andquiescing time were choosen as the main facts of baicalin extracting, L9 (34) orthogonal experimentaldesign was done,and the baicalin content was used as an index of searching for the best preparing process.The study indicated that the optimum extraction process as follows: adding 10 times and 8 timesamount of water for the first time and the second time respectively, contenting of tween-80 was0.5%, incubation time of acid praecipitatum was 1.5h, and the quiescing time was 3h. Theexperiment was repeated three times in this way, the content of baicalin was 11.38%. The densityand content of alcohol, the time and frequencies of backstreaming were seen as main facts oflumbricus extracting, being selected with the L9 (34) orthogonal experimental design as thelumbricus content index, which was another essential component in this praeparatum, searching forthe best preparing process. The study indicated that the optimum extraction process as follows: the amountof alcohol was 8 times, the density of alcohol was 70%, extracting once at 90℃, the time ofextracting was 3h, The experiment was repeated 3 times follow this preparing process, thelumbricus content was about 33%in dry extraction. Other chinese traditional herbs exceptscutellaria and lumbricus were extracted by decoction and alcohol sedimentation technique, and preparedfor physic liquor which contained 1g/ml of the crude drugs, and then mixed with the extraction ofscutellaria and lumbricus. Some additives were added into the praeparatum to ensure the qualityand stability, and the category and dosage were also tilted. The result indicated that natriumbenzoicum was the best antiputrefactive and the content of which was 0.15%; As the pH was 5.5~6.5,the clarity was best, and the liquor could suspend well-distributed and keep stab.In the study of the quality control criteria, the TLC was used to identify the scutellaria, honeysuckble and licorice, the separation of the stains was apparent and the negative contrast wasnot disturbed. All of these proved that the method being used was specific. HPLC was used todetermine the amount of baicalin, the condition of chromatographic analysis of baicalin was asfollow: diamonsil C18 column (4.6mm×150mm,5μm), the mobile phase consisted ofmethanol-water-glacial acetic acid (50:49:1) at a flow-rate of 1.0 ml/min, the ultraviolet detectorwas set at 335nm, the column temperature was at the room temperature, the sample size was 10μl.All of the results showed that the preparing technology was rational, the quality of this medicinewas sTab and the operation was convenient. So a best method was used as the basis to explore thetentative stability of baicalin of Manhukang oral liquor. When the amount of caffeotannic acid wasused as the index, HPLC was used to determine the amount of caffeotannic acid. The condition ofchromatographic analysis of baicalin was as follow: diamonsil C18 column(4.6mm×150mm,5μm),the mobile phase consisted of AN-water-glacial acetic acid (15:100:0.15) at a flow-rate of 1.0ml/min, the ultraviolet detector was set at 326nm, the column temperature was the roomtemperature, the sample size was 10μl. The results showed that, in this condition, the averagerecoveries of caffeotannic acid in the test solution were good. The resolving power between thechromatographic peak of caffeotannic acid and others exceeded 1.5, the negative control wasnon-interference. So a best method was gained which was the basis of exploring the tentativestability of caffeotannic acid of Mnahukang oral liquor.Then the affecting factors including indoor temperature, accelerating experiment were studied,all of the results showed that the preparing technology was rational and the quality of this medicinewas sTab.In conclusion, Mnahukang oral liquor was made of the extract if chinese traditional herbs, itwas safe, effective and could be used in clinic for long time. In the meanwhile, the quality could becontrolled effectively by modern scientific technique. This study would provide the foundations ofcreating more social benefit and economic profits.
