Study On The Preparing Technique Of Longyin Oral Liquid

Objective: We studied and prepared the Longyin oral liquid, optimized the preparation technique, to establish high performance liquid chromatography assay of Longyin oral liquid initially.Methods:(1) The prescription of Longyin includes Gentiana, Artemisia, Gardenia, Bupleurum, and Glycyrrhiza. We used the orthogonal experiments to select the best extraction process parameters of Longyin oral liquid, according to the evalution factors which contained extraction time, extraction times and quantity of the solvent times, based on the extract yield and amount of Gentio picroside.(2)The contents of layer chromatography was used for the qualitative identification of Gentiana, Artemisia, Gardenia, and Bupleurum in prescription, to establish the quality control method of Longyin Oral LiquidResults:(1) The final condition of the extraction process is acceding to ten times the amount of water to extract time is 2 h, and extracting for 2 times;(2) The linear range of Gentiopicroside was 0.06 ~ 0.30 mg/m L( 0.6 ~ 3.0 μg), Y = 13168178.3333 x-54651.3000(r = 0.9999,n = 5),the average recovery of Gentiopicroside was 101.2%, RSD was 1.56%(n = 6). Thin layer chromatography indicated that Gentiana, Artemisia, Gardenia, and Bupleurum could show that the spot separation effect is good. According to the measured results of multiple samples, A initially content of the standard was setup, for each 1 m L Longyin oral liquid contains Gentiopicroside respectively is not less than 4.0 mg.Conclusions:(1) The research results of the Longyin oral liquid preparation process showed that the preparation technology is stability and reliable, easy to operate, small cost and suitable for mass production.(2) This study to create high performance liquid chromatography method had the characteristics of high sensitivity, precision, easy to operate, the product quality is stable, and can be controlled, which provided practical basis for the quality standards of Longyin oral liquid.
