Rose (Floribunda Roses) is one of perennial and wood flowers, which belongs to therosaceous of rose and is one of the most famousest flowers in our country. It is usually called thequeen flower. It is loved by world wild people because of its excellence like variousvariety, colourful like, long florescence etc. It plays an important role in folwer industry.The radiation breeding as an important breeding method is always taken by the largenessbreeding workers. 60Co radiation can enhance the mutational frequency, promote generecombination and the successful hybridization of distant strain. Using the RAPD can detect thevariation degree of the mutation's genome in the molecular level. It also can appraise and choose themutations of radiation descendant early through the anaysis on relativity between the genomevariation degree and radiation dose, enhance the breeding effficiency. So it has the important theoryand practice significance.This experiment confirmed the suitable radiation dose of the the tissue culture's rose plantthrough the tissue culture in connection with 60Co-γinduced mutation,analyzed the productionways and variation character of the M1 generation and the relativity between the radiation dose andcoefficient of variation of the M1 generation through the RAID. The primary content and result areas follows:(1) It is not suitable to choose the too small plant when we confirmed the tissue culture's roseplant as the plant material in the radiation breeding. We should choose the adult plant that haverooted and about 10cm high to carry on the radiation treatment.(2) The semi-lethal of the tissue culture's rose plant (LD50)is about 50Gy. It is not suitable thatthe largest radiating dose surpasses 110Gy. The experimental result indicated that the plants diedcompletely when the radiation dose surpasses 110Gy.(3) when we uesed the tissue culture's rose plant as the plant material in the radiationbreeding, the production ways of variated branch and variated plant were mainly three kinds: thevariated plant sprouted from the root when the upside plant's growth came under the suppression;the variated branch sprouted from the plant after the plant growing point died;: the variated plantsprouted from the root after the upside plant died.(4) The variated of variated branch and variated plant mainy represented in shape of leaf andtrunk, antidisease.The shape of leaf variation represented that the leaf changde to be smaller and partite; The shape of trunk variation represented that the plant changde to be lowed andcompact; antidisease variation is chance variation, we found 2 plants that represented antidiseasevariation in M1 generation;(5) We observed the shape of the stoma and palisade tissue though the electronmicroscope, found that the stoma of M1 is larger and the form of-palisade tissue is tightnesserexcept 1plant than CK.(6) The radiation dose had an extremely positive relativity with variation coefficient of theM1 generation. We can appraise and choose the mutations of radiated descendant early in the themolecular level using the method of RAPD, enhance the breeding efficiency.
