Z. tenuifolia is a warm-season grass has excellent characteristics, such asadaptability, strong drought resistance, the expansion of power and trampling of strongresistance and wear resistance, high elasticity properties, and so on. When the soil andclimatic conditions were suitable, for the general fertility management, Z. tenuifolia couldform dense, neat lawns which can be widely used in the courtyard lawn, golf courses,airports, stadium and other urban green belt. However, there also existed weaknesses, likeslower germination under natural conditions or sprouting, often need trimming, wastehuman and financial resources, the long time does not prune easy to produce the grass pitand the litter of weeds and so on.This experiment was for the purpose of the characteristics of wild Z. tenuifoliaresources and its biological characteristics and turf quality evaluation, like morecomprehensive experimental study, the testing research for the grass resourcesdevelopment and utilization of wild Z. tenuifolia provided some references. On the basisof discussing disinfection and treatment the explants in the plant regeneration andselection hormone for induction callus, adjustment the callus condition in the subcultureand methods of plant regeneration to establish an efficient system of renewable tissueculture. Combined traditional breeding methods with modern biotechnology to shorten thevariety breeding cycle, and laid a solid foundation for the genetic engineering andsomaclonal variation study. Test results of this research were as follows:1. The wild grass collected from the northern of Thailand was identified as the wildgrasses of Z. tenuifolia, named for "taiyin 1" provisionally;2. Quality Evaluation of "taiyin 1" grass was great;3. In this test the optimal combination of explants sterilization was 1 minute alcoholtreatment, 0.1% mercuric chloride for 8 minutes. The contaminated rate was 7.43%,browning rate was 8.95%, the Survival was 75.72%;4. The best medium for callus induction was MS+3.0 mg/L 2,4-D+0.1 mg/L6-BA+IAA 1.0 mg/L, healthy callus induction rate has reached 67.13%; 5. The best medium for callus subculture was MS+1 mg/L 2,4-D+0.5 mg/L BA,suitable for adjustment of callus status;6. The best medium for callus tissue differentiation was MS+2.0 mg/L KT+3.0 mg/LBA+0.2 mg/L NAA, the differentiation rate reached 67.38%;7. The rooting medium was 1/2MS+0.2 mg/L NAA, the highest rate of rooting was58.82%.
