The Research On Tissue Culture And Propagate Technology Of Wild Lily In 'Qinba' Mountains

In order to propagate and preserve the wild lily in'QinBa'Mountains effectively, the tissue culture study use wild'YiChang'lily and lilium lancifolium thumb in'QinBa'Mountains as materials. We separately studied how to obtain test-tube plantlet, the various factors which effect bulblet enlarge in vitro and transplant the plantlet. The results as follows:1. Tissue culture of wild'Yichang'lilyThe results showed that the appropriate medium on which the adventitious buds could be induced directly from scales was MS+BA 2.0 mg/L+NAA 0.05 mg/L.The ability of scales make adventitious buds as Down>Middle>Upper;The suitable medium on which the buds could be induced from leaves was MS+BA 1.0 mg/L+NAA 0.1 mg/L+2,4-D 2.0 mg/L.The Induction rates of adventitious bud were Down was high,Middle and Upper were low;The optimum proliferation medium for adventitious buds was MS+BA 1.0 mg/L+NAA 0.05 mg/L.The appropriate medium for rooting was 1/2 MS+ NAA 0.4 mg/L+AC 1.0 g/L. After this study we know that there are obvious rule about inducing adventitious buds with different parts of scales and leaves.2. The study on formation and enlargement of bulblet in vitroMainly research on the effect of photoperiod,temperature,paclobutrazol and the proportion and total quantity of NH4+and NO3- in culture medium during bulblet formating and enlarging in vitro.The findings indicted that different period of darkness did have a significant effect on the weight and diameter of bulblet in tube, The two treatments, 12 h/d and 16 h/d darkness,had better effect; The rate of sucrose utilized in culture medium under different photoperiod were different and the best darkness treatment was 16 h/d. 22℃is suitable for bulblet enlarge;Rate of effective weight under 28℃is the best one.When temperature is 22℃the effect of rate of sucrose utilized in culture medium is best.The suitable paclobutrazol concentration is 12 mg/L.When NH4+/NO3- is 30/40,the fresh weight and the rate of effective weight are better than others.When the total nitrogen quantity is 70 mM,weight of bulblet is the best one in all treatments.3. The transplantation of bulblet Fertilizing 1/2MS nutrition which add 10ml (NH4)2SO4(10.31 mM) is favorable for the growth of plantlet.The bulblets which formate by paclobutrazol have better effect than bulblets which formate by nitrogen treatment.The former have wider leaves,the latter have more number of leaves and more higher.
