Pepper(Capsicum annuum L.) belongs to Solanacea family in Capsicum spp., itis the most important vegetable crop in acreage after Chinese cabbage in China now.Heterosis in pepper are quite extensive and apparent, the production of using excellenthybrid seeds is larger than the conventional ones by 30%.So 90% of commercialproduction pepper seeds are hybrid seeds now. Pepper hybrid seeds production usingmale sterile could lower cost by reducing time and labour, increase the genetic of theF1 seeds and protect the intellectual property. So the breeders are more and moreinterested in researching the physiological and biological of cytoplasmic malesterile(CMS) in pepper. This experiment was studied on the location analysis offertility restoration major gene in CMS pepper. The reaction systems of AFLP andRAPD in pepper were optimized by adjusting the key factors in the reaction systems.Two markers which tightly linked with the fertility restoration major gene inchromosome 6 were located in this 2 000 segregated population. Some markers linkedwith the fertility restoration major gene which had located were proved in thissegregated population. Hot pepper which has strong restore fertility ability crossedwith sweet pepper. In order to prove that if can transfer the restoration gene throughthe hot pepper to sweet pepper in molecular level. The main research results arefollows:1 The reaction systems of AFLP and RAPD in pepper were optimized.2 Two markers: E32M49-424bp and q18-1.0kb which tightly linked with thefertility restoration major gene in chromosome 6 were located.3 By using the Joinmap 3.0 software and depended on the restore fertilitycondition of the segregated population,the distance between AFLP markerE32M49-424bp and fertility restoration major gene is about 3.1 cM;The distancebetween RAPD marker q18-1.0kb and fertility restoration major gene is about 2.2cM.4 It proved that SCAR marker Rf-737 linked with the fertility restoration majorgene. The distance between SCAR marker Rf-737 and fertility restoration major geneis about 12.7 cM;The distance between SCAR marker Rf-737 and AFLP markerE32M49-424bp is about 15.8 cM;The distance between SCAR marker Rf-737 and RAPD marker q18-1.0kb is about 10.5 cM.5 It proved that RAPD marker L09- 763 had no special polymorphism diversityin the segregated population.6 Hot pepper(xiangtanwan) which has strong restore fertility ability crossed withsweet pepper(83-60),in the F1 generation had found the special polymorphismdiversity.
