Molecular identification and genetic diversity of Malus hupehensis (Pamp.) Rehd.(Tea Crabapple) accessions which maintained at Malus hupehensis (Pamp.) Rehd.germplasm nursery of Fruit Tree and Tea Institute of Hubei Agriculture Academyusing 17 SSR makers which developed and screened from apple (Malus domesticaBorkh.), in an attempt to provide molecular level evidence for germplasm collection,maintenance, and the origin and migration research of Malus hupehensis (Pamp.)Rehd.. The 33 accessions herein come from six populations in the west of Hubeiprovince and one population in Linyi, Shandong province. Results of SSR analysisshowed:1. High levels of genetic diversity were detected with a mean of 7.12 alleles perlocus, a mean percentage of polymorphic loci P=98.3, and a mean expectedheterozygosity He=0.76.2. Each of primers except GD15, GD162, Z71981 could differentiate in all 33accessions with 6 genotypes. 'Xingshan' genotype included all 18 accessionsgenerated from 'Xingshan' population. 'Jianshi No.4' and 'Longwan No.3' accessionown the same genotype to 4 accessions come from 'Fangxian' population named'Fangxian' genotype. Each two accessions come from 'Hefeng', 'Xuanen' ,'Longwan' has unique genotype, respectively. 'Pingyitiancha' has a unique genotypealso. Obviously, Malus hupehensis germplasm nursery held many duplicateaccessions.3. UPGMA cluster analysis of all accessions based on NTSYS 2.10 gave rise to adendrogram which was divided into two groups at genetic similarity (GS) 0.63. Firstgroup consist of 'Xingshan' and 'Hefeng' genotype, GS=0.71, while 'Fangxian','Xuanen', 'Longwan' and 'Pingyitiancha' cluster second group.4. Results of ploidy analysis showed that no accessions herein was diploid. Theorigin of Malus hupehensis (Pamp.) Rehd. was challenging to unravel.
