In this research, based on the DH population with 119 lines derived from theNo.2127-17 (yellow-seed parents) and Zhongyou 821 (black-seeded parents), themensurations of seed coat color were compared, a genetic linkage map of Brassica napusL. was constructed and the QTL of the seed coat color trait was analyzed. The mainconclusions are as follows:1. By comparison of the two mensurations of seed coat color, namely seed coatpigments distill analysis and UltraScan XE mensuration, both of the mensurationscan evaluate the seed coat color. The process of seed coat pigment distill analysiswas complex, didn't fit for the mensuration of populations material. Themanipulation of UltraScan XE mensuration was contrast easy and the results wereaccurate. Four parameters of seed coat color could get once, the efficiency washigher.2. Analysis the DH population genotypes with SSR markers and based on theLOD=4.0, a genetic linkage map was constructed by using JionMap 3.0, which wasmade up of 23 linkage groups, including 162 SSR markers, covering 834.1 cM intotal with an average interval of 5.15 cM between adjacent loci. As for the differentlinkage groups, the length varied from LG9 of 3.2 cM to LG1 of 141.9 cM. Theaverage marker interval distance varied from 1.6 cM (LG9) to 9.83 cM (LG9).3. The seed coat color of DH populations was mensurated by UltraScan XEmensuration and one marker analysis method of QTL Cartographer V2.5 wasemployed in QTL analysis of seed coat color trait. A total of seven QTLs (qScc1—qScc7) were detected on five linkage groups with LOD=3.0, namely two on LG1,each one on LG2, LG3, LG10, LG13 and LG16, respectively. Phenotypic variationexplained by single putative QTL varied from 14.95% to 77.14%.4. Phenotypic variation of the major QTL (qScc6) on LG13 varied from 32.15% to77.14%. It links with the SSR marker Na14-E08, which is the one linked withyellow-seeded gene from Xiao's study, the interval space is 10.6 cM. The other SSRmarker Nal2-E02 from Liu's QTL study on seed coat color links with qScc1, qScc2and qScc5, its interval space is 17.8 cM, 7.5 cM and 1.6 cM, respectively.
