| This study on the cytology of Lamprotula fbrosa consist three parts of experiments, The first experiement studied on the type of the hemocytes by using three different staining techniques and two different obtain techniques. The second experiment studied on the karyotype of LamprotuIa forosa by using the gill tissue. The third part was a primary culture experiment, the hemocytes, gill tissue and mantle tissue were culrured. The purpose of this experiment is getting to know the basic conditions of a sucessful primary culture, at last the study compared with this three experiments to value the their advantages and inferiors.1. The hemocytes of Lamprotula fibrosa was classified into four maior categories: granulocyte, agranulocyte, hyalinocyte and lymphocyte-like cell. The study confirmed the exits of intermediate cell.2.The study confirmed that the suitable pH for the primary culture of Lamprotula fibrosa was 6.9, and the suitable medium program for all the three types of cell. The result of primay culture showed that the speed of hemocytes adhere on the glass were the fastest; The speed of gill tissue adhere were medium; The speed of mantle tissue adhere on the glass were slow. The multiply circles of hemocytes, gill tissue, mantle tissue were 6days, 10days and 11days.3. The diploid number ofLamprotula fibrosa was 2n=38, The karyotye formula was 2n=24m+6sm+8st, NF=68. The karyotype contains 12 pairs of metacentric, 3 pairs of submetacentirc and 4 pairs of subtelocentric chromosomes. There is no telocentric chromosome at all. |
