An endogenic activator protein isolated from Altemaria was a high performance, wide-spectrum, non-toxicity to human beings and livesyocks, non-pollutional and novel inducer. The activator protein could activate a series of responses, and activate seed germination and growth and induce immunoreaction to a large number of diseases of plant by binding to the acceptor of cell membrane. In this study, growth status of tomato seedling, the dynamic change of some enzymes related to growth of seedling, such as dehydrogenase in root. In this study, the systematic dynamic characteristics of phenylanine ammonia-lyase (PAL), superoxide dismutase (SOD)and catalase (CAT) were studied in tomato leaves which were inoculated with the Alternaria solani and treated with different concentrations of plant activator protein. The results are listed as follow:1. In this study, it screened activator protein from four pathogenic fungi of different genus, including Alternaria solani (Ell. et Mart), Alternaria alternata (Fries) keissler, Alternaria alternata (Fries) keissl, Fulvia fulva (Cooke) Cif. And then these four kinds of activator protein were applied to seeds treatment of tomato, pepper, cabbage, cucumber. Results showed that the activator protein of Alternaria solani had the best activity in promoting the average germination and average root length of the four vegetable seeds.2. Effect of activator protein from Alternaria solani on the average germination ratio, average bud length, root lengh and plant height of tomato seedlings was studied. Results showed that growth of tomato seedling was obviously promoted when the crude activator protein was diluted by 10-3 times and sprayed on the seedling leaves, while the crude activator protein with high concentration inhibited tomato seedling growth. However, there was not so much difference when the activator protein was diluted by 2×10-3 times and 6.67×10-4 times.3. Effect of activator protein from Alternaria solani on dehydrogenase activities was studied: Treated with activator protein, dehydrogenase activity of tomato seedlings increased and was still higher than that of the control after 40 days, respectively. Especially, when the concentration were 10-3 times, the effect was the most significant, reached the most than that of the control, and its lasting time was the longest.4. Effect of activator protein from Alternaria solani on resistance to Alternaria solani and induce resistance of tomato seedling: After being treated with activator protein, direct resistance to Alternaria solani was not observed.5. Effects of plant activator protein from Alternaria solani on tomato leaves of important defense enzymes were detected: (1) After being treated with Alternaria solani and plant activator protein, plant activator protein can induce the expression of these defense enzymes respectively in tomato leaves for PAL,CAT and SOD. The optimum concentration of plant activator protein is 10-3 times. (2) The activity of PAL and SOD in tomato leaves were higher than the control meanwhile CAT activity was decreased. The results indicated that and the induction of PAL and CAT expression in susceptible tomato cultivar Da-yu was generally lower than in resistant tomato cultivar Qian-fu, The results showed that the induction of PAL, SOD expression in tomato leaves was positively related to tomato resistance to Alternaria solani, and could be used as the indicators of tomato resistance to Alternaria solani.
