| This article takes the dual hybrid piglet of da bai and the chang bai as the reaserching aim of different days of age from 0d,1d,2d,3d,4d,to 35d in order to study the condition and function of the FcRn expression in the gastrointestinal tract of piglet,it applies the RT-PCR and the radioimunologlcal technique to the primary discussion of the distribution rule of the levels of FcRn Receptor and IgG of blood serum on the bisis of the dual hybrid piglets of different day of age in the gastrointestinal tract.1.Applying RT-PCR to detecting the level of FcRn in thegastrointestinal tract of piglet among different periods,the result is shown below:The expression c ondition of FcRn mRNA in the different tissues of the gastrointestinal tract of piglet is shown below:FcRn mRNA expresses in all parts of the gastrointestinal tract of piglet; the expression of FcRn is high in the duodenum and jejunum,the highest value of FcRn/GAPDH (IOD)is up to 0.87;the expression of FcRn is low in the colon and stomach,the lowest value of FcRn/GAPDH(IOD)is 0.3.It shows that the.mainpart of the FcRn to转è¿the IgG in the colostral milk rather than stomach and colon.The expression condition of FcRn mRNA of the different day of age of the piglet in different gastrointestinal tract is shown below:the FcRn mRNA in different tissues expresses stably of the 0dpiglet without feeding colostral milk,the quantity of expression is lower than the 0d piglet without feeding colostral milk but higher than the piglet after 7d,the difference of all parts except duodenum and jejunum is not conspicuous in the first seven days;the highest expression of FcRn mRNA in the gastrointestinal tract of piglet is on 0d and 1d;the difference of duodenum and jejunum is conspicuous on 0d,1d and after 7d;it gradually lowered after 14d,the difference is not conspicuous in the first 4days,the expression of FcRn lowered very much on 7d except that the difference of stomach and colon in contrast to other tissues;the lowest value of FcRn in the gastrointestinal tract of piglet is on 28d,i.e.afteræ–奶.The rules above shows that the genetic factor of FcRn expression is predominant,the high density of IgG enhances its expression to some extent.The piglet obtains the maternal source IgG mainly in the first four days and especially the first two days. 1.Applying the ELISA to detecting the level of IgG in the blood serum of piglet of different periods,the result is shown below:The IgG cannot be detected in the blood serum of the 0d piglet without feeding colostral milk, which proves that the piglet cannot obtain the maternal source antibody during the period of fetus and it can only obtain the maternal source antibody through feeding thecolostral milk. The IgG in the blood serum of piglet rises step by step from 0d to 4d,therefore,it reaches the highest point on 4d,then gradually lowered,it reaches the lowest point on 21d,then gradually heightened,which proves that the IgG originated from milk from birth to 21d,after 21d the IgG mainly depends on the production of its self-immunity.The intestinal tract of piglet mainly depends on the FcRn to transport the IgG from milk to blood.The IgG in the piglet lowers with the time increasing,then the expression of the FcRn of piglet,which makes IgG in the body of piglet lower.The study indicates that two days after birth the piglet makes FcRn transport IgG in the colostral milk through duodnum and jejunum,it thus obtains the maternal source antibody. |
