In this study, total RNA was isolated from tissues of Tibetan and Taihu pigs, then,using RT-PCR method to proliferate H-FABP gene and the sequences of products wereanalyzed. Using GAPDH as internal standard control, the regular of H-FABP gene mRNAexpression levels was investigated by semi-quantitative RT-PCR. Longissmus muscle ofTibetan and Taihu pigs was sampled to analyze developmental changes of intramuscular fat(IMF) content by Soxhlet method. The results are as follows:1. The coding sequence (CDS) of Tibetan and Taihu pig H-FABP gene was 402bp.Between AJ416019 (GenBank Accession Number) and Tibetan pig and Taihu pig, thehomology rates of nucleotide sequences of coding region of H-FABP gene ware 99.8% and100% respectively. By analysis of nucleotide mutation sites in coding region of H-FABPgene, there was a transition as T→C in Tibetan pig, it existed on site of 152. There was nomutation site in Taihu pig. The cDNA sequences of H-FABP gene have been deposited inGenBank database, the accession numbers are EF619344 and EF622095 for Tibetan andTaihu pig respectively.2. In longissmus muscle of Tibetan pigs, H-FABP mRNA expression level was thelowest at the age of 2 months, reached the highest at the age of 6 months, and then declinedcontinuously at the age of 8 and 12 months. The expression level at the age of 8 and 12months showed no differences. In longissmus muscle of Taihu pigs, H-FABP mRNAexpressed the highest on birth, and a little low at the age of 1 and 3 months. There was noexpression at the age of 2, 4 and 5 months.3. Expression level of H-FABP gene mRNA in four different tissues from Tibetan andTaihu pigs at the age of 2 months were analyzed. As to Tibetan pigs, expression level wasthe highest in adipose. Expression level in heart was higher than in longissmus muscle andin longissmus muscle was higher than in liver. As to Taihu pigs, expression level was thehighest in adipose. Expression level in heart was higher than in liver. There was noexpression in longissmus muscle.4. The IMF content increased continuously with growing and showed highly significant different (P<0.01)between ages in Tibetan pigs. As to Taihu pigs, IMF contentincreased dramatically from birth to month 1, and declined dramatically from month 1 to 2,then increased continuously from month 2 to 5. Except month 2 with 3 (P>0.05), IMFcontent in different months showed highly significant different (P<0.01).
