Gram-negative bacteria infection was the most common bacteriosis on veterinary clinic. These bacterium were able to release an endotoxin in the process of antibiosis curing besides they had pathopoiesia self, and an endotoxin can bring out uncontrolled systemic inflammatory response syndrome (SIRS) and septic shock, cause multiple organ failure (MOF) and clinical death. But we still did not adopt the method to "Treating for bacterium and endotoxin simultaneous/y", and there still were not the relative medicines. We designed Dan-Fu Injection with Florfenicol and Sa/via mi/tiorrhiza Bunge, and had set up a method of detection Dan-Fu Injection by HPLC, and on the basis of studying in the pre-prescripting, the orthogonal design was used to optimize the prescripttion with three factors, which included pH value, mixture solvent, antioxidants, and three levels of them .And we studied the stability and the quality standard of Dan-Fu injection. At last, we did the Pharmacodynamics experiments, which included the antibacterium activities of Dan-Fu Injection ex vivo, the effects of SOD activities and MDA level after administration Dan-Fu Injection in the plasma of toxemic piglets infected swine choTera salmonella, the curative effect of Dan-Fu Injection curing piglets infected swine cholera salmonella. The result was indicated:1. The method which detected Florfenicol, Salvianic acid A and Prtocatechual dehyde in Dan-Fu Injection by HPLC was stable, reliable, and its repeatability was good, its recovery was good, its result was accurate, so it can be used to detect contents of Dan-Fu Injection. 2.The optimum prescription was that N,N-Dimethylacetamide and Propylene glycol were solvent, and N,N-Dimethylacetamide' s capacity was 55%, Vitamin E was antioxidants and its concentration was 0.3% , pH was 6.0.3. Dan-Fu Injection was stabile under illumination (4500 Lx) ; and it showed instability under the high temperature (40℃) condition; Its appearances and colour, pH value, clarificaty can reach a request in the long-term testing, and Florfenicol was stabile, but Salvianic acid A and Prtocatechualdehyde' s stability remained to be improved.4. The study of pharmacodynamics experiments indicated that Dan-Fu Injection was sensitive to swine cholera salmonella, chicken bacillus coli, swine bacillus coli separated from clinical and rabbit bacillus coli separated from clinical; it can restore SOD activity and MDA level in plasma of toxemic piglet, which indicated it was able to resist free radicals and endotoxin further; its curative effect that cured piglet infected swine cholera salmonella by man was good: piglet' s temperature restored quickly. The clinical symptom disappeared quickly. The liver, lung and spleen' s pathology damage restored well.
