Four mating types of basidispores in 3 strains of Pleurotus ostreatus weredetermined using a nuclear migration test and an OWE-SOJ technique. The nuclearmigration test showed that nuclear migration only occurred in A=B≠mating interactionbut did not in A≠B= interaction. In OWE-SOJ test, a fluffy colony without clampconnection formed in the A=B≠mating interaction where nuclear migration occurs, whilea barrage colony without clamp connection formed in A≠B= interaction where nuclearmigration do not occur. The two colonies could be distinguished clearly from each other.The result of the nuclear migration test is consistent with that of OWE-SOJ, which couldbe corroborated each other. Therefore it is feasible to apply the two to accuratedetermination of mating types in Pleurotus ostreatus. The polymorphism of genomicDNA in spore monokaryons and protoplasted monokaryons derived from two dikaryoticstrains, Huaping49 and No. 1 Florida Var. and their crossing progenies was analyzed byRAPD technique. By analysis of the RAPD profiles each spore monokaryon withdifferent mating types was easily differentiated one another. In addition, the two crossingcombinations(T1×T2, T3×T4) also clearly were distinguished each other and from thedikaryotic parental strains.
