Cultivation spawns used were solid ones mostly in China currently. Reciprocally liquid spawns were fewer used as cultivation spawns and a little only used as second spawns.The chief reason was that liquid spawns could not store for long time.There were more advantages of liquid spawns than that of solid spawns. Present research explored the solidification technique to prolong the storage time of liquid spawns.The most cultivation quantity of mushroom in China was oyster mushroom.So Pleurotus ostreatus Tianda 300,which was cultivated widely in Henan province,was selected as experiment material.Firstly,the optimum liquid medium for Tianda 300 growth was chosen from 6 liquid media by shaking flask.The main components were corn powder,bean cake powder,sucrose,and yeast powder.In this medium the mycelium pellet wet weight of Tianda 300 was high to 0.18kg/L after fermentation 7.5d,it was much higher that in the other 5 media.The pilot fermentation test in 40L fermenter showed that the size of mycelium pellets were suitable and wet weight was high to 0.12kg/L after cultured for 3.5d.Two methods for liquid spawn solidification were performed.The method of mycelium pellets mixed with carrier media was superior to the other one of fermentation broth mixed with carrier media.The solidification spawn maked by mycelium pellets and the medium group constituted with cottonseed shell,maize core,and bran,could germinate after inoculated 48h,and growth rate was 0.49cm/d,as well as the surviving rate of cultivation bags was more 80%.It was superior to the other 3 carrier media groups.The optium proportion range ofmycelium pellets to cartier media was 1:6~10.The physiological characteristic parameters including breath intensity,cellulase activity,and isozymograms of esterase,and polyphenol oxidase and peroxidase of solidification spawns stored for 0d to 60d period were analyzed.The results showed that the breath intensity,isozymograms of esterase,and peroxidase gradually increased, but isozymograms of polyphenol oxidase steadily weakened and lastly disappeared during the store period,Otherwise the hypha growth rate of the solidification spawn stored for 15d was the highest and the change model of cellulose activity was "M"style during the store period.The fuitbody yield of solidification spawns was not different with the normal spawns,it indicated that the life cycle of solidification spawns could prolong to 60d efficiently.The estimate cost of solidification spawns was about 0.755 yuan RMB/kg and lower than that of normal spawns.So solidification spawns were worthy to practice using.
