Antagonism, esterase isozyme electrophoresis, ITS and ISSR analyses, combing with cultivation experiment, were employed on the taxonomic identification and genetic diversity of 61 cultivated Pleurotus spp. strains from Henan province. The results are as follows:1. According to antagonism reaction, tested strains can be divided into 13 vegetative compatibility groups,the other strains have the same name with the 13 strains, the most one are 18.2. 20 strains were ISSR augmented using 8 primers and 106 loci were amplified. The percentage rate of polymorphism loci (P) was 95.4%. The number of observed alleles (Na) was 2.0000, the number of effective alleles (Ne) was 1.7844, the Nei's gene diversity (H) was 0.4368 and Shannon's information index (I) was 0.6279 in every loci. The Nei genetic distance (D)is 0.00~0.60 between starins calculated through NTSYSpc 2.10e software package, The dendrogram was constructed based on cluster analyzing using UPGMA method.3. The esterase isozyme electrophoresis analysis showed that the esterase isozymograms of these 13 chosen strains are different and of ample polymorphism, while the isozymograms of the non~antagonism strains showed no difference.4. The ribosome Internal Transcribed Spacer 1 of the 13 chosen strains were amplified with the primer of ITS1 and ITS4, and about 700bp segments were obtained. The results of sequencing and blasting experiments showed that 1 strain is Pleurotus ostreatus, 4 strains are Pleurotus florida, 1 strain is Pleurotus pulmonarius and 7 strains are Pleurotus conucopinus.5. According to the cultivation experiment, the tested strains can be classified into 4 species: Pleurotus ostreatus, Pleurotus pulmonarius, Pleurotus florida and Pleurotus conucopinus.It was summarized that the oyster mushroom cultivated in Henan province are Pleurotus ostreatus, Pleurotus pulmonarius, Pleurotus conucopinus and Pleurotus florida, which could provide strain with clear germplasm in production; there is obvious discrepancy among the tested strains through Cluster Analysis using esterase isozyme, ISSR and ITS method, which indicated that there is rich genetic diversity of oyster mushroom strains in Henan province.
