Study On The Main Biological Characters Of Hibiscus Moscheutos L.

Hibiscus moscheutos L.is studied on biological characters,cytology characters, Verticillium dahliae resistances,genetic relationship between Hibiscus moscheutos L.and different cotton species.Through the study of botanic characters and stress resistance,the Floras about Hibiscus moscheutos L.are perfected,the potential value of Hibiscus moscheutos L.to improve cotton genetic sources is confirmed farther,the information about Hibiscus moscheutos L.genetic resources is more systematicness,and cotton genetic resource research is done between genera;The chromosome number of Hibiscus moscheutos L.is ensured,the karyotype analysis and the meiotic process of pollen mother cell of Hibiscus moscheutos L.are researched,which provide cytological basis for the later worker about Hibiscus moscheutos L.and cottons;The genetic relationship among Hibiscus moscheutos L.and cottons was estimated at molecular level(combined morphological markers).The main results were as follows:1.When the flowers of Hibiscus moscheutos L.were opened just,the pollen viability is strong;the life-span of the pollen may reach three days at the room temperature;the proper condition of the pollen bourgeon is pH5.0,32℃,the concentration of the cane sugar 30% and the concentration of the boric acid 1 mg/L～10 mg/L;the diameter of the microspore is 123.76μm.Hibiscus moscheutos L.is the herbaceous perennial plant,it has two froms of produce which are sexual propagation and asexual propagation;the flowers open in morning and close at night,self-sterile but the fruit-setting rate is very low.2.The well-spread and clear mitotic metaphase chromosomes of Hibiscus moscheutos L. were obtained,2n=2x=38,N=19.The karyotype analysis indicated that there was a pair of satellites on chromosome No.1,and the average length of Hibiscus moscheutos's chromosomes was 5.96±0.83μm.The arm ratio of 10.5%chromosomes was over 2,and the ratio of the longest chromosome to the shortest one was 1.68.The karyotype was 2A and the karyotype formulae was 2n=2x=38=32m+6sm(2SAT).No abnormal behavior was observed in the meiotic process of pollen mother cell of Hibiscus moscheutos.There were nineteen bivalents(19Ⅱ)at metaphaseⅠ,indicating it was a typical diploid species.3.Hibiscus moscheutos L.can high resistant level which induced by Verticillium dahliae in two ways of greenhouse and asepsis-table at seedling stage.Verticillium dahliae cultivated can't be separated from the disease plants of Hibiscus moscheutos L.in greenhouse and asepsis-table. 4.The DNA of 6 materials genome is amplified with 57 SSR primer pairs,23 SSR primer pairs can amplify in the DNA of 6 materials genome,120 locis are detected totally,average 5.2 loci per primers.6 materials can be classified into two groups 2 at 0.08,the genetic relationship of cotton isⅠclass,Hibiscus moscheutos L.isⅡclass,by itself.
