From the study of plant growth regulators and their combination, base media, darkening treatment and other factors which affect on the stem subculture and proliferation, callus inducing, regenerating and rooting, we developed a regeneration system of stem , leaf and hypocotyls which from Armeniaca sibirica (L.) Lam.(1) Stem of Armeniaca sibirica (L.) Lam grown best on MS+6-BA 1.0 mg/L+IBA 0.2 mg /L. Stem clusered with higher 6-BA which was 2.0mg/L and grown much callus at the bottom of the plants with higer IBA which was 0.3mg/L. Proliferation multiples and≧0.5cm shoot rates were the highest when the adventitious shoots were cultured on MS+6-BA 0.8 mg/L+IBA 0.2 mg/L. After15 days roots grown in succession on MS IBA 0.4mg/L .The rooting rate was 86.7%,average root number was 3.8 and the largest root number was 5-6.Finally, regeneration plants from stem of Armeniaca sibirica (L.) Lam have been got.(2) The callus were found at the petiole and vein of leaves after 10d in dark, and adventitious shoots were found after 40-60d exposured to light. MS+TDZ 2mg/L+ NAA 0.75mg/L+ AgNO3 4.0mg/L was better for callus inducing rate and regeneration rate, which were 100% and 11.1% respectively. The best result was obtained when the leaf adaxial side was touching the culture medium. In this position the regeneration obtained was 25.1%, which was higher than when the leaf abaxial side was in contact with the medium.(3) After 15d in dark and then exposed to light ,the rate of shoots regenerated was 16.6% and callus induced was 100% when cultured on the half NH4NO3 of MS medium supplemented with TDZ 2.0mg/L and NAA 0.5mg/L. A relative higher callus inducing rate(97.2%) and regeneration rate(13.8%) were also get when 6-BA 1.0mg/L and NAA 0.5mg/L was used. TDZ was better than 6-BA in plant regeneration. The effect of basal medium was modified MS > QL > WPM.
