The Research Of Optimizing The System Of In Vitro Maturation Of Oocytes And In Vitro Culture Of Zygotes To Xu-huai White Goats

In this paper, the influence of heparin, season of oocytes collected and different cell factors on in vitro maturation (IVM), in vitro fertilization (IVF) and zygotes in vitro culture (IVC) of Xu-huai white goat oocytes were studied, the main results were as follows:1. The BO fluid which was added 10μg/mL heparin for Xu-huai white goat sperm in vitro Capacitation was the best advantageous for in vitro fertilization.2. Compared with October ~January, more available oocytes could be gathered from Xu-huai white goat ovaries in March~June.The season had a significant influence (P<0.05) on Xu-huai white goat oocytes IVM, the rate of maturation was higher in March~June, but there was no significant influence on oocytes IVF (P>0.05).3. It could be effectively enhance the rate of maturation, rate of 2-cell (P<0.01) and the rate of blastula (P<0.05) with adding the hormone to the mature fluid,the group of added 10 unit/mL FSH, 30 unit/mL LH and lμg/mL E2 was the best.4.There was a significant difference on the rate of 2-cell, 4~16-cell and blastula between control group and the groups added different density of GDNF, LIF or SCF to the mature and zygote fluids (P<0.01). The group added LIF was better for early embryo growth than the groups of adding SCF or GDNF. In the whole, the groups added 15 ng/mL GDNF, 10 ng/mL LIF or 10 ng/mL SCF were the better. 5. Three kind of cell factors had the synergism, could promot Xu-huai white goat oocyte to in vitro mature and the embryo early growth. The groups added 10 ng/mL LIF, 10 ng/mL SCF or 10 ng/mL GDNF to the mature and zygote fluids could effectively enhance the rate of oocytes maturation and the early embryo growth potential.
