The Genetic Diversity Analysis Of Melting Peach (Prunus Persica) Germplasms In ZheJiang Province

We used thirty-one microsatellites dispersing on the international peach genetic linkage maps(T×E)to study the genetic diversity of sixty-two peach cultivars collected and selected in Zhejiang province.A total of 88 polymorphic alleles were generated.The number of alleles per locus ranged from 2 to 5 alleles,with a mean of 2.8 putative alleles per locus.The observed heterozygosity is from 0.02 to 0.89,with a mean of 0.48 per microsatellite; the expected heterozygosity is from 0.06 to 0.73,with a mean of 0.49;the shannon index(Ⅰ)is from 0.46 to 1.29.,with a mean of 0.77.Seven highly polymorphic microsatellites were selected our from these microsatellites,they are CPPCT029, CPPCT040,BPPCT001,UDP96-013,UDP96005,BPPCT025 and CPPCT025.A total of 30 polymorphisms were produced.The number of alleles per locus ranged from 4 to 5 alleles with a mean of 4.29 putative alleles per locus.We can identify unambiguous the 62 peach cultivars with 7 pairs of SSR primers.We obtained specific fingerprints with highly polymorphic SSR primers in this research.These printfingers can use to identify the cultivares.The primer CPPCT029 can distinguish 'zaohualu' cultivars,'yanhong' cultivars from other cultivars,primer Pchgms 6 for 'bafanbaitao',primer CPPCT040 for 'yanhong' cultivars,primer UDP96-013 for 'zaotaimi' cultivar;BPPCT028 can separate 'xiangtu' cultivars, 'zaohualu' cultivars,'xinchuanzhongdao' cultivars from other cultivars;UDP97-401 can distinguish 'juwangtao' cultivar from other cultivars;BPPCT025 can distinguish 'datuanmilu' cultivar from other cultivars,CPSCT006 can also distinguish 'zaohualu' cultivar from other cultivars.We can diverageⅠ,Ⅱ,ⅢandⅣfour cluster for Sixty-two peach cultivars (selections)using UPGMA method at the similarity coefficient 0.69.There are twenty peach cultivars in clustersⅠ,mostly introduced from Japan,included 'Hakuho','hujingmilu' cultivars.There are ten peach cultivars in clustersⅡ,mostly come from Anhui province,including 'zhongqiumi','laishanmi' cultivars The clusterⅢis small,it only contain the 'yanhong' cultivars and 'cangfangzaosheng' cultivars and other cultivars belong to clusterⅣ.In this cluster,most are the local peach cultivars:'Yulu' peach cultivars and their progeny.The peach cultivars in Zhejiang province can be always grouped together,it can explain these cultivars rarely cross with peach from other provinces.To some extent,these conclusions can apply theretical inferences on the analysis of peach genetic diversity.
